HPLC法测定复方连翘膏改良制剂中黄芩苷的含量

Determination of baicalin in optimizational compound Lianqiao cream by high performance liquid chromatography

目的:建立复方连翘膏改良制剂中黄芩苷的高效液相色谱含量测定方法。方法:色谱柱为Agilent Extent C_(18)柱(250 mm× 4.6 mm,5 μm),甲醇-乙腈(1:1)为A相,0.4%磷酸水溶液为B相,A相-B相(35:65)等度洗脱,流速1 mL·min^(-1),检测波长280 nm,柱温30℃,进样量10μL。结果:黄芩苷在2.37-94.98μg·mL^(-1)范围内线性关系良好(r=1.0000);黄芩苷的平均回收率为97.65%,RSD为1.25%(n=9)。结论:本方法专属性强、灵敏度高、重现性好,可用于复方连翘膏改良制剂中黄芩苷的含量测定,为改良制剂的质量控制提供简便可靠的测定方法。

Objective:To establish a method for the determination of baicalin in optimizational compound Lianqiao cream by high performance liquid chromatography(HPLC).Methods:The chromatographic column was Agilent Extent C_(18)(250 mm × 4.6 mm,5 μm).The methanol-acetonitrile(1:1)was phase A,0.4%hydrophosphate solution was phase B,phase A-B(35:65)was eluted equally.The flow rate was 1 mL·min^(-1).The detection wavelength was 280 nm.The column temperature was set up at 30℃.And the injection volume was 10 μL.Results:Baicalin had a good linear relationship in the range of 2.37-94.98 μg·mL^(-1)(r=1.0000).The average recovery of baicalin was 97.65%with the relative standard deviation(RSD)of 1.25%(n=9).Conclusion:This method was specific,sensitive and reproducible,which was suitable for the determination of baicalin in optimizational compound Lianqiao cream,and provided a simple and reliable assay method for the quality control of the improved preparation.
