链终止测序技术在RNA结构分析中的应用

摘要 双脱氧链终止法是最常用的DNA测序技术,其核心是依赖双脱氧核糖核苷酸的扩增终止技术。在课堂授课中,教师对于双脱氧链终止法的原理及应用讲解较少,学生普遍对于该方法理解不足。基于以上问题,在核酸化学的教学内容上引入SHAPE(Selective 2’-Hydroxyl Acylation and Primer Extension)技术,不仅取得了较好的教学效果,同时增强了学生对于科研的兴趣。对SHAPE技术的原理及结果分析等教学设计简要叙述,以期为核酸化学的教学提供建议与参考。 Dideoxy sequencing method is the most common method of DNA sequencing technology,which was dependent on dideoxy ribonucleotide.However,the principle and application of dideoxy sequencing method is also seldomly involved in the classroom teaching,and the students are deeply lacking understanding.Based on the above issues,SHAPE technique was added into teaching content.That not only get good feedback on teaching,but also can enhance students’interests in research.This paper will briefly describe the dideoxy sequencing methods which could provide reference for the teaching on nucleic acid.
