肝细胞癌组织TROAP表达与上皮-间质转化的相关性

Correlation between expression of TROAP and epithelial-mesenchymal transition in hepatocellular carcinoma tissues

目的探究肝细胞癌(hepatocellular carcinoma,HCC)患者癌组织中Trophinin相关蛋白(Trophinin-associated protein,TROAP)的表达情况,并分析TROAP与上皮-间质转化(epithelial-mesenchymal transition,EMT)的关系。方法选取在成都市第三人民医院行根治性肝癌切除术的97例HCC患者,于术中收集患者HCC组织及癌旁组织,基于GEPIA数据库分析HCC组织及正常组织中TROAP的表达情况及其表达与HCC患者预后的关系,采用免疫组化法检测TROAP蛋白、E-钙黏蛋白(E-cadherin,CDH1)及波形蛋白的表达,分析TROAP蛋白表达与HCC患者临床病理特征的关系,采用Spearman法分析TROAP蛋白与CDH1、波形蛋白表达的相关性,Kaplan-Meier法分析TROAP蛋白表达情况与患者生存率的关系。结果HCC组织中TROAP蛋白及波形蛋白的阳性率高于癌旁组织(P<0.05),CDH1蛋白阳性率低于癌旁组织(P<0.05)。TROAP蛋白阳性率与HCC患者的Edmondson分级、TNM分期、淋巴结转移及肿瘤结节数目有关(P<0.05),且TROAP蛋白表达与CDH1表达呈负相关(P<0.05),与波形蛋白表达呈正相关(P<0.05)。TROAP蛋白阳性表达者累积生存率明显低于阴性表达者(P<0.05),与GEPIA数据库结果一致。结论HCC组织TROAP蛋白阳性率高于癌旁组织,TROAP表达与EMT过程有关,有望成为判断HCC患者预后的潜在标志物。

Objective To explore the expression of Trophinin-associated protein(TROAP)in the cancer tissues of patients with hepatocellular carcinoma(HCC),and to analyze the relationship between expression of TROAP and epithelial-mesenchymal transition(EMT).Methods Ninety-seven patients with HCC who underwent hepatectomy in Chengdu Third People′s Hospital were selected,and HCC tissues and adjacent tissues of the patients were collected during the operation.The expression of TROAP in HCC tissues and normal tissues of patients and the relationship between its expression and the prognosis of HCC patients was analyzed based on the GEPIA database,the expression of TROAP protein,E-cadherin(CDH1)and vimentin was detected by immunohistochemical method,the relationship between TROAP protein expression and clinicopathological characteristics of HCC patients was analyzed,the correlation between TROAP protein and CDH1 and vimentin expression was analyzed by Spearman method;the relationship between TROAP protein expression and survival rate of patients was analyzed by Kaplan-Meier method.Results The positive rates of TROAP protein and vimentin in HCC tissues were higher than those in adjacent tissues(P<0.05),and the positive rate of CDH1 protein was lower than that in adjacent tissues(P<0.05).The positive rate of TROAP protein was related to the Edmondson grade,TNM stage,lymph node metastasis,and the number of tumor nodules in HCC patients(P<0.05),and the expression of TROAP protein was negatively correlated with the expression of CDH1(P<0.05),and positively correlated with the expression of vimentin(P<0.05).The cumulative survival rate of patients with positive expression of TROAP protein was significantly lower than that of patients with negative expression(P<0.05),which was consistent with the results of the GEPIA database.Conclusion The positive rate of TROAP protein in HCC tissue is higher than that in adjacent tissues.The expression of TROAP is related to the EMT process and it is expected to be a potential marker for judging the prognosis of HCC patients.
