不明原因稽留流产绒毛组织E-CAD/CK7/PLAC8表达

EXPRESSION OF E-CAD/CK7/PLAC8 IN CHORION TISSUE OF UNEXPLAINED MISSED ABORTION

目的探讨E-钙黏蛋白(E-CAD)、细胞角蛋白7(CK7)和胎盘特异性蛋白8(PLAC8)在不明原因稽留流产绒毛组织中的表达及其可能的意义。方法选取早孕期不明原因稽留流产病人20例(稽留流产组)、非计划妊娠行人工流产的正常早孕女性20例(正常妊娠组)作为研究对象,采用实时荧光定量聚合酶链反应(qRT-PCR)检测绒毛组织中E-CAD、CK7及PLAC8的mRNA表达,Western Blot及免疫组织化学方法检测绒毛组织中E-CAD、CK7及PLAC8的蛋白表达。结果稽留流产组绒毛组织中E-CAD、CK7的mRNA和蛋白表达均明显高于正常妊娠组(t=3.995~12.200,P<0.01),两组PLAC8的mRNA和蛋白表达差异无显著性(P>0.05)。E-CAD以及CK7的阳性染色主要定位于滋养层细胞的细胞膜,PLAC8的阳性染色主要定位于合体滋养层细胞的细胞核,稽留流产组绒毛组织中E-CAD、CK7的平均光密度值(AOD)明显高于正常妊娠组(t=3.823、3.889,P<0.05),两组PLAC8的AOD差异无显著性(P>0.05)。结论绒毛组织E-CAD、CK7表达升高可能与不明原因稽留流产的发生有关。

Objective To investigate the expression of E-cadherin(E-CAD),cytokeratin 7(CK7),and placental-specific protein 8(PLAC8)in chorion tissue of unexplained missed abortion and their possible significance.Methods A total of 20 patients with unexplained missed abortion in early pregnancy were enrolled as missed abortion group,and 20 women with normal early pregnancy who underwent induced abortion due to unplanned pregnancy were enrolled as normal pregnancy group.Quantitative real-time PCR was used to measure the mRNA expression of E-CAD,CK7,and PLAC8 in chorion tissue,and Western Blot and immunohistochemistry were used to measure the protein expression of E-CAD,CK7,and PLAC8 in chorion tissue.Results Compared with the normal pregnancy group,the missed abortion group had significantly higher mRNA and protein expression le-vels of E-CAD and CK7 in chorion tissue(t=12.200,9.968;P<0.01),and there was no significant difference in the mRNA and protein expression levels of PLAC8 between the two groups(t=8.447,P>0.05).The positive staining of E-CAD and CK7 was mainly localized in the cell membrane of trophoblast cells,and the positive staining of PLAC8 was mainly localized in the nucleus of syncytiotrophoblast cells.Compared with the normal pregnancy group,the missed abortion group had a significantly higher average optical density(AOD)of E-CAD and CK7 in chorion tissue(t=3.823,3.889;P<0.05),and there was no significant difference in the AOD of PLAC8 between the two groups(P>0.05).Conclusion Increases in the expression of E-CAD and CK7 in chorion tissue may be associated with the occurrence of unexplained missed abortion.
