摘要
目的 研究醒脑静注射液对急性酒精中毒大鼠的促醒作用及氧化应激损伤的保护作用及机制。方法 将大鼠随机分成空白组、模型组和醒脑静注射液低、中、高剂量组(1.5、3.0、6.0 mL/kg),模型组及各给药组给予52%乙醇造模,待翻正反射消失后静脉注射生理盐水或醒脑静注射液。记录翻正反射消失时间;试剂盒测定肝脏乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)、超氧化物歧化酶(SOD)活性以及丙二醛(MDA)、谷胱甘肽(GSH)含量;实时定量PCR检测肝组织中ADH、ALDH mRNA表达量。结果 与模型组相比,醒脑静注射液中、高剂量组翻正反射消失时间明显缩短(P<0.01),肝脏ADH、ALDH、SOD活性及ADH、ALDH mRNA表达水平、GSH含量显著提高(P<0.01),但MDA含量无明显差异(P>0.05)。结论 醒脑静注射液能提高ADH、ALDH的活性促进乙醇代谢,对急性酒精中毒大鼠有促醒作用,同时可提高SOD活性和GSH含量,减少酒精所致氧化应激对肝脏的损伤。
Objective:To study the effect and mechanism of Xingnaojing Injection(XNJI)on the wake-up and the protective effect of oxidative stress injury in rats with acute alcoholism. Methods:The rats were randomly divided into blank group,model group and XNJI low,medium and high dose groups(1.5,3.0 and 6.0 mL/kg). The model group and each drug group were given 52% alcohol. After righting reflex disappeared,XNJI or saline was injected intravenously. The loss of righting reflex(LORR) was recorded;the activities of alcohol dehydrogenase(ADH),aldehyde dehydrogenase(ALDH),superoxide dismutase(SOD) and the contents of malondialdehyde(MDA),glutathione(GSH)in liver tissue were detected by kit;the mRNA expressions of ADH and ALDH in liver tissue were detected by real-time quantitative PCR. Results:Compared with the model group,the LORR in the medium and high dose XNJI groups were significantly decreased(P< 0.01),and the activities of ADH,ALDH,SOD and the content of GSH and the m RNA expression levels of ADH and ALDH in the liver were significantly increased(P< 0.01),but there was no significant difference in the content of MDA(P >0.05). Conclusion:XNJI can improve the activities of ADH and ALDH,promote the metabolism of alcohol,promote the wake-up of rats with acute alcoholism,increase the activity of SOD and the content of GSH,and reduce the damage of liver caused by alcohol induced oxidative stress.
