摘要
目的探讨下腔静脉腹主动脉夹闭在大鼠创伤性脑损伤后脑组织在体心脏灌流固定术中的应用效果。方法自由落体打击诱导中度创伤性脑损伤24 h后,股静脉穿刺置管静脉注射伊文斯兰染色,对照组行左心室心脏灌流,实验组左心室心脏灌流同时夹闭下腔静脉胸主动脉。断头取脑常规HE染色及免疫组化染色。结果实验组灌注固定成功率为93.33%,高于对照组灌注固定法的80.00%,实验组灌注时间(102.25±4.12)s,对照组灌注时间(248.53±4.10)s,差异有统计学意义(P<0.05)。实验组灌注液使用量(103.85±3.12)mL,对照组使用量(148.12±3.16)mL,差异有统计学意义(P<0.05)。两种方法灌注固定损伤区蓝染,光镜下星形胶质细胞肿大,神经元核固缩。结论与对照组比较,实验组灌注成功率高,灌注时间缩短,灌注液使用减少,节约试剂,且可以取得与传统方法固定脑组织相同的良好效果。
Objective To investigate the effect of abdominal aorta clipping of inferior vena cava on intracardiac perfusion after traumatic brain injury in rats.Methods Twenty-four hours after the moderate traumatic brain injury induced by free fall,the femoral vein was catheterized and intravenous injected with Evans’blue staining.The control group received left ventricular cardiac perfusion,and the experimental group received left ventricular cardiac perfusion while clipping the decapitated inferior vena cava thoracic aorta for routine HE staining and immunohistochemical staining.Results The success rate of perfusion fixation in the experimental group was 93.33%,which was higher than that in the control group(80.00%).The perfusion time in the experimental group was(102.25±4.12)s,while that in the control group was(248.53±4.10)s,and the difference was statistically significant(P<0.05).The amount of infusion used in experimental group was(103.85±3.12)mL and that in the control group was(148.12±3.16)mL,and the difference was statistically significant(P<0.05).The injured area was stained blue,and the astrocytes were enlarged and the neuronal nuclei were pyknotic under lignt microscope.Conclusion Compared with the control group,the experimental group has a higher success rate of perfusion,shorter perfusion time,less use of perfusion fluid,saving reagents,and can achieve the same good effect as the traditional method of brain tissue fixation.
