摘要
为探究脂联素及其受体基因在绵羊中的序列结构及组织表达特异性,以兰州大尾羊为研究对象,利用RACE技术克隆兰州大尾羊脂联素及其受体基因的全长cDNA序列,运用实时荧光定量PCR技术分析脂联素及其受体基因在兰州大尾羊15种组织中的表达规律。结果表明:兰州大尾羊APN、AdipoR1和AdipoR2基因的cDNA全长分别为2101 bp、2035 bp和1089 bp,APN基因在肌肉、心脏及皱胃中的表达量显著高于其他组织,AdipoR1和AdipoR2基因在内脏脂肪与皮下脂肪中的表达量显著高于其他组织。APN和AdipoR1基因在大肠、小肠、瘤胃及皱胃中均有一定程度的表达,而AdipoR2基因在其余组织中几乎不表达。
In order to explore the sequence structure and specificity of tissue expression in adiponectin and its receptor gene in sheep,Lanzhou fat-tailed sheep was selected as research object,the full-length cDNA sequence of adiponectin and its receptor genes in Lanzhou fat-tailed sheep were cloned by using rapid amplification of cDNA end(RACE).The real-time fluorescent quantitative PCR(qPCR)was used to analyze the expression patterns of the adiponectin and its receptor gene in 15 tissuesof Lanzhou fat-tailed sheep.The results showed that the full-length cDNAs of Lanzhou fat-tailed sheep APN,AdipoR1 and AdipoR2 genes were 2101 bp,2035 bp and 1089 bp,respectively.The expression of APN gene in muscle,heart and abomasum was significantly higher than that in the other groups,while the expression of AdipoR1 and AdipoR2 genes in visceral fat and subcutaneous fat was significantly higher than thatin the other groups of Lanzhou fat-tailed sheep.In addition,APN and AdipoR1 genes were expressed to a certain extent in the large intestine,small intestine,rumen and abomasum,while AdipoR2 genes were almost not expressed in the other tissues.
