番红花花蕊分化期差异蛋白表达的鉴定与分析

摘要 通过蛋白质组学筛选和研究番红花花蕊分花期差异蛋白的变化,旨在揭示番红花花蕊分化的内在分子调控机制。利用MALDI/TOF/TOF-MS/MS质谱鉴定与分析技术筛选与鉴定番红花花蕊分化期的差异蛋白,结果表明:75个差异表达蛋白得以鉴定,分析其可能的功能后构建了番红花花蕊分化的分子调控模型,其中,糖代谢途径为分化提供了足够的能量和营养;氨基酸代谢中的甲硫氨酸tRNA连接酶、精氨酸酶等可能与细胞分裂素、多胺等分子信号调控有关;活性氧代谢中的抗坏血酸过氧化氢酶、过氧化氢酶、硫氧还蛋白过氧氢酶、甘露糖-3′5′-异构酶等可能调控植物激素信号和抗性机制;热激蛋白、蛋白伴侣保障蛋白修饰和加工的正常进行;延伸因子Tu、β-肌动蛋白、β-微管蛋白、3-oxo-γ(4,5)-类固醇-5-β-还原酶构建细胞结构。本研究鉴定了75个可能在番红花花蕊分化中发挥重要作用的蛋白,并分析了其可能的调控过程,为进一步揭示番红花花蕊分化分子调控机制提供了科学依据。 In order to explore the molecular regulation mechansim on the flower bud differentiation of Crocus sativus L.,protein abundance changes were studied by using approach of proteomics to screen the different proteins.MALDI/TOF/TOF-MS/MS identification and analytical techniques were used to compare and analyze the changes of the proteome for the clarifying the biological functions of key proteins during the process of differentiation.The results showed that 75 differentially expressed proteins were reliably identified,and their metabolism model was preliminaryly constructed after their possible function was analyzed.The glycolytic pathway provided sufficient energy and nutrition for differentiation;methionine tRNA ligase,Arg,etc.in amino acid metabolism might be related to the regulation of molecular signals,such as CTK and PA;APX,CAT,TpxⅡ,GME.in ROS metabolism,might regulated the phytohormone signals and maintained the resistant mechanisms,and HSP,chaperone ensured the normal progress of protein modification and processing;3-oxo-γ(4,5)-steroid-5-β-reductase,EF-Tu,β-actin,β-tubulin constructed cell structure.The identified differential proteins that maybe play important role and regulate possible differentiation process provide a comprehensive reference to understand molecular mechanism of flower bud differentiation in Crocus sativus L.
