半枝莲多糖组分抗肿瘤作用和免疫调节的实验研究 被引量:6

摘要 目的:探讨半枝莲多糖组分的抗肿瘤作用,并对其免疫调节作用进行研究。方法:首先提取半枝莲多糖组分,然后以H22肝癌移植瘤小鼠为模型,设置模型组(等量生理盐水)、黄芪多糖组(100 mg/kg)和半枝莲多糖组分(SBP-2A)低、中、高剂量组(25、50、100 mg/kg),每天给药1次,连续给药12 d,比较各组抑瘤率、脏器指数、γ-干扰素(IFN-γ)及瘤组织蛋白表达水平指标,评价半枝莲多糖组分对H22荷瘤小鼠肿瘤生长及免疫功能的影响。结果:与模型组相比,半枝莲多糖组分(SBP-2A)低、中、高组瘤体质量均明显减少(P<0.05),对H22实体瘤抑制率分别为15.42%、38.67%、29.95%;与模型组对比,半枝莲多糖组分(SBP-2A)低、中、高组小鼠脾指数和胸腺指数均升高;血清中IFN-γ水平显著增高(P<0.05),瘤体组织中STAT1蛋白表达水平显著上调(P<0.05),且呈半枝莲多糖组分浓度剂量依赖性关系;半枝莲多糖组分(SBP-2A)高剂量组与模型组STAT1蛋白表达水平比较,有统计学意义(P<0.05)。结论:半枝莲多糖组分对H22荷瘤小鼠具有抑瘤作用,其作用可能与调节机体免疫功能有关。 Objective:To explore the anti-tumor effect and the immunomodulatory effect of Scutellaria barbata polysaccharide.Methods:Firstly,the polysaccharide of Scutellaria barbata was extracted,and then the H22 liver cancer transplanted tumor mice were used as models and divided into the model group(with the same amount of normal saline),the Astragalus polysaccharide group(100 mg/kg)and the Scutellaria barbata polysaccharide groups of Low-dose(25 mg/kg),medium-dose(50 mg/kg)and high-dose(100 mg/kg).The intervention was once a day for 12 consecutive days,and the tumor inhibition rate,organ index,interferon-γ(IFN-γ)and tumor tissue protein expressions were compared in each group,to evaluate the effects of Scutellaria barbata polysaccharide on tumor growth and immune function in H22 tumor-bearing mice.Results:Compared with the model group,the tumor mass was significantly reduced in Scutellaria barbata polysaccharide groups(P<0.05),and the tumor inhibition rates were 15.42%,38.67%and 29.95%,respectively.Compared with the model group,the spleen index and the thymus index of the mice were increased,and the serum level of IFN-γwas also significantly increased in the Scutellaria barbata polysaccharide groups(P<0.05),which showed dose-dependent effect.The protein expression of STAT1 in tumor tissue was significantly up-regulated in the high-dose group compared with that in the model group(P<0.05).Conclusion:The polysaccharide of Scutellaria barbata have anti-tumor effect on H22 tumorbearing mice,which may be related to the regulation of immune function.
