摘要
为了探讨甘草酸能否降低雷公藤多苷治疗胶原诱导性关节炎(collagen-induced arthritis,CIA)所致的血液系统毒性作用,采用牛Ⅱ型胶原蛋白免疫诱导Wistar大鼠建立CIA动物模型,CIA大鼠随机分为模型组、雷公藤组和配伍组,另取未造模鼠作为对照组,每组8只;雷公藤组和配伍组灌胃给药;模型组和对照组给予等量灭菌生理盐水,处理6周.影像学、HE及Masson染色评判大鼠关节及软骨病理变化情况、外周全血血常规细胞计数、Giemsa染色法观察骨髓细胞数目及形态变化、HE镜下观察脾脏组织病理变化.结果显示,与CIA模型组相比,雷公藤组大鼠血液中红细胞计数显著降低(P<0.05),骨髓粒系、红系细胞减少;配伍组红、白细胞计数与雷公藤组相比,大鼠血液中红细胞计数明显增加(P<0.05),并且骨髓有核细胞数目增多.据此,可知甘草酸配伍雷公藤多苷能够降低单独应用雷公藤多苷治疗胶原诱导性关节炎所致的血液系统毒性作用.
In order to investigate whether glycyrrhizic acid(GA)can reduce the hematologic toxicity of tripterygium glycosides(TG)in the treatment of collagen-induced arthritis(CIA),a CIA animal model was established by injecting typeⅡcollagen into Wistar rats.The CIA rats were randomly divided into a model group,a TG group and a compatibility(TG+GA)group,and rats without injection were taken as the control group.Each group contained 8 rats.The TG group and the compatibility group acquired intragastric administration while the model group and the control group were given equal amounts of sterilized physiological saline.After six weeks,medical images,HE and Masson staining were used to evaluate the pathological changes of rats joints and cartilage,peripheral blood routine cell count and Giemsa staining method to observe the number and morphological changes of bone marrow cells,and HE microscope to observe the pathological changes of spleen tissue.The results showed that compared with the CIA model group,the red blood cell count of the TG group significantly decreased(P<0.05),and the myeloid and erythroid cells in bone marrow decreased.Compared with the TG group,the red blood cell count and white blood count of the compatibility group increased significantly(P<0.05),and the number of bone marrow nucleated cells also rose.According to the above results,we can draw a conclusion that GA can reduce the hematologic toxicity of TG in the treatment of CIA.
