摘要
为提高甘薯脱毒种苗无糖培养效率,以甘薯脱毒种苗作为试验材料,建立无糖培养体系,设计单因素试验和正交试验,对试验数据进行单因素方差分析、正交试验直观分析与方差分析,进一步优化无糖培养技术。结果表明:带有根系的甘薯脱毒种苗进行无糖培养效果较好;最优的无糖培养液成分为MS(有机物)+0.1 g/L花宝2号+0.67 g/L抑菌剂;最优的室内培养条件:培养基质为泥炭土与珍珠岩(1∶1),培养液体积为1 200 mL,选择3根28 W日光灯(光照强度约为2 900 lx),光照时长为12 h/d。
In order to improve sugar-free culture efficiency of sweet potato virus-free seedling, virus-free Dioscorea esculenta(Lour.) Burkill seedlings were used as experimental materials, sugar-free culture system was established, single factor experiment and orthogonal experiment were designed, and the experimental data were analyzed by single factor variance analysis, orthogonal experiment visual analysis and variance analysis to optimize the culture conditions of sugar-free culture technology. The results showed that the sweet potato virus-free seedlings with roots had better sugar-free culture effect. The optimal sugar-free medium was MS(organic) + 0.1 g/L Huabao 2 + 0.67 g/L bacteriostatic agent, and the optimal indoor culture conditions were as follows: the culture medium was peat soil and perlite(volume ratio 1∶1), the volume of culture solution was 1 200 mL, three 28 W fluorescent lamps(light intensity was about 2 900 lx) were selected, light duration was 12 h/d.
