miR-150、Beclin-1及ELOVL6在原发性痛风患者外周血单个核细胞中的表达及与病情的关系研究

摘要 目的 分析miR-150、Beclin-1及ELOVL6在原发性痛风患者外周血单个核细胞(PBMC)中的表达及与病情的关系。方法 收集2018年3月至2021年5月我院收治的原发性痛风患者98例(GA组),根据患者临床症状分为急性期(AGA组)47例,非急性期(NAGA组)51例;另选取同期健康体检者84例(对照组)。对比不同人群中miR-150、Beclin-1、ELOVL6、尿酸(UA)表达情况;AGA组与NAGA组miR-150、Beclin-1、ELOVL6、UA、视觉模拟评分(VAS)表达情况,分析miR-150、Beclin-1及ELOVL6与UA、VAS评分相关性。结果 GA组miR-150、Beclin-1、ELOVL6蛋白表达低于对照组,UA表达高于对照组(P<0.05);AGA组miR-150、Beclin-1、ELOVL6表达低于NAGA组,UA、VAS评分高于NAGA组(P<0.05);miR-150、Beclin-1及ELOVL6与UA、VAS评分之间均为负相关(P<0.05)。结论 miR-150、Beclin-1及ELOVL6在原发性痛风患者PBMC中的表达均下调,与患者病情进展关系密切。 Objective To analyze the expressions of miR-150, Beclin-1 and ELOVL6 in peripheral blood mononuclear cells(PBMC) of patients with primary gout and their relationship with the disease.Methods A total of 98 patients with primary gout admitted to our hospital from March 2018 to May 2021 were selected(GA group). According to the clinical symptoms of the patients, they were divided into an acute phase(AGA group, n=47) and a non-acute phase(NAGA group, n=51). In addition, 84 volunteers undergoing physical examination in our hospital during the same period were selected as a control group. The expressions of miR-150, Beclin-1, ELOVL6, and uric acid(UA) in different populations were compared. The expressions of miR-150, Beclin-1, ELOVL6, UA, and visual analogue scale(VAS) were compared between the AGA group and the NAGA group. The correlation between the levels of miR-150, Beclin-1 and ELOVL6 and the UA and VAS scores was analyzed.Results The expressions of miR-150, Beclin-1 and ELOVL6 were significantly lower while the expression of UA was significantly higher in the GA group than those in the control group(P<0.05). The expressions of miR-150, Beclin-1 and ELOVL6 were lower while the scores of UA and VAS were higher in the AGA group than those in the NAGA group(P<0.05). The expressions of miR-150, Beclin-1 and ELOVL6 were negatively correlated with the UA and VAS scores(P<0.05).Conclusions The expressions of miR-150, Beclin-1 and ELOVL6 in the PBMC of patients with primary gout are all down-regulated, and are closely related to the patient′s disease progression.
