摘要 肾脏进展性纤维化是各种慢性肾脏病进展的重要原因,其病理特征是肾间质的成纤维细胞大量活化增殖,过度合成和分泌细胞外基质。低氧诱导因子-1α(HIF-1α)是使机体适应缺氧环境的关键调节因子。在慢性肾脏病纤维化发展过程中,HIF-1α发挥着重要作用。本文对HIF-1α的结构、生物学功能及其在肾脏纤维化中的作用进行综述,探索以HIF-1α信号为靶点的治疗对肾脏病及其并发症的潜在价值。 Progressive renal fibrosis is an important cause of the progression of various chronic kidney diseases.Its pathological characteristics are massive activation and proliferation of renal interstitial fibroblasts,excessive synthesis and secretion of extracellular matrix.Hypoxia-inducible factor-1α(HIF-1α)is a key regulator in adapting the body to hypoxia.HIF-1αplays an important role in the development of fibrosis in chronic kidney dis ease.This article reviews the structure,biological function and role of H IF-1αin renal fibrosis,and explores the potential value of HIF-1αsignaling targeted therapy for renal disease and its complications.
