多元醇和糖的HPLC-RID检测方法优化及其在四种饮料酒中的应用

摘要 利用高效液相色谱-示差折光检测法(High-performance liquid chromatography-refractive index detector, HPLC-RID)同时测定7种多元醇和糖。优化后该方法最适色谱分析条件是示差折光检测器温度为33℃、流动相为乙腈-水(95∶5,V/V)、色谱柱温度为30℃和流速为0.6 m L/min;且7种多元醇和糖的混合标准品在0.1~10 mg/m L浓度范围内呈良好的线性关系(R2>0.9952),检测限范围为0.019~0.058 mg/m L,定量限范围为0.043~0.098 mg/m L,方法精密度的相对标准偏差(RSD)为1.37%~2.39%,加标回收率为83.66%~112.70%。对饮料酒的分析结果显示,黄酒主要含有果糖,配制酒、露酒和果酒根据原辅料的不同或含不同比例的木糖醇、阿拉伯糖、果糖、赤藓糖醇和甘油。此方法为不同种类饮料酒的质量评价提供了方法借鉴。 High-performance liquid chromatography with a refractive index detector(HPLC-RID) method was used for the simultaneous determination of seven polyols and sugars. After optimization, the optimum chromatographic analysis conditions were 33℃ for the refractive index detector cell temperature, acetonitrile-water(95:5, V/V) as the mobile phase, 30℃ for the column temperature, and 0.6 m L/min for the flow rate. Moreover, the seven polyols and sugars mixed standards in the concentration range of 0.1~10 mg/m L were in a good linear relationship(R2>0.9952), the limits of detection were within the range of 0.019~0.058 mg/m L, the limits of quantitation were within the range of 0.043~0.098 mg/m L, the relative standard deviations(RSDs) of method precision were 1.37%~2.39%, and the recoveries were 83.66%~112.70%. The analysis results showed that huangjiu mainly contained fructose. depending on the raw ingredients, integrated alcoholic beverages, lujiu, integrated and fruit wines contain different proportions of xylitol, arabinose, fructose, erythritol and glycerol.This method would provide a methodological reference for the quality assessment different types of alcoholic beverages.
