摘要
探讨金银花总黄酮含量的最佳提取工艺,对其体外抗氧化活性进行研究,初步研究金银花总提取液和主要功效成分绿原酸的抗氧化活性。以总黄酮得率为指标,分别以水和乙醇进行提取,选择较佳提取溶剂,在此基础分别进行浸提和索式提取,选择较优提取方式,采用单因素和正交实验法优化金银花总黄酮的提取工艺,同时对金银花提取液和绿原酸的体外抗氧化活性进行研究。实验结果表明金银花总黄酮的最佳提取工艺为乙醇浸提,料液比1:15,乙醇浓度70%,温度70℃下提取2 h。体外抗氧化实验结果金银花提取液具有显著的抗氧化能力、还原能力及羟自由基和DPPH自由基清除能力,相同浓度条件下金银花提取液的抗氧化活性高于绿原酸的抗氧化活性。研究结果为金银花资源的开发利用提供理论依据。
The optimum extraction process of total flavonoids content in Flos Lonicerae was discussed, and its antioxidant activity in vitro was studied. The antioxidant activity of chlorogenic acid and Flos Lonicerae extract had been preliminarily studied. Flos Lonicerae was respectively extracted by water and ethanol to choose the better extraction solvent with flavonoids yield as the index. On this basis, the better extraction method of Flos Lonicerae was confirmed between leaching extraction and soxhlet extraction. Using single factor experiment and orthogonal experiment optimized the extraction technology of total flavonoids in Flos Lonicerae, and at the same time, the in vitro antioxidant activity of Flos Lonicerae extract and chlorogenic acid were studied. In vitro antioxidant activity of Flos Lonicerae extractive and chlorogenic acid had been studied. The experimental results show that the optimum extraction technology of total flavonoids from Flos Lonicerae for ethanol extraction, and solid-liquid ratio 1:15, 70% ethanol concentration, extraction of 2 h under 70 ℃ temperature. In vitro antioxidant experiment result, that Flos Lonicerae extractive solution has remarkable antioxidation ability, reducing capacity, hydroxyl radical and DPPH free radical scavenging ability, and the antioxidant activity of the Flos Lonicerae extractive solution under the same concentration condition is higher than the antioxidant activity of chlorogenic acid. The results of this study provide theoretical basis for the exploitation and utilization of Flos Lonicerae resources.
