胱氨酸基荧光碳点的制备及其黄体酮的荧光恢复响应

摘要 将胱氨酸(Cys)碱性溶液在恒温水热反应后,可制得荧光碳点(Carbon Dots, CDs),添加柠檬酸(CA)为碳源后,形成的氮硫掺杂碳点CA-Cys-CDs具有较高的荧光量子产率和较好的光稳定性。实验表明,该碳点对Pb^(2+)离子有特异性响应,碳点溶液的荧光信号可被Pb^(2+)猝灭。pH=5.72的B-R缓冲溶液条件下,在CA-Cys-CDs-Pb^(2+)体系中加入黄体酮,可使体系的荧光信号得以恢复。在9.90×10^(-7)-1.07×10^(-5)mol/L浓度范围内,体系的荧光强度恢复程度与黄体酮浓度呈现良好的线性关系,线性方程为I/I_(0)=5.83×10^(4)c+1.02,相关系数为0.9908,检出限为3.7×10^(-7)mol/L。将该方法用于黄体酮注射液中黄体酮含量的检测,所测结果为药品标示量的93.2%-112%。 Fluorescent carbon dots(CDs) were synthesized in this study with the alkaline solution of cystine as the primary raw material by thermostatic hydrothermal reaction.The experimental results showed that N,S-dopped CDs(CA-Cys-CDs) have been synthesized assisted by citric acid(CA) with high photo-stability and quantum yield.Due to the nitrogen and sulfur co-doped structure on the surface of CA-Cys-CDs, the carbon dots had a specific response to lead ions with fluorescence quenching.In the B-R buffer solution of pH=5.72,when introducing progestin into the CA-Cys-CDs-Pb^(2+)solution, the fluorescence signal recovered by blocking the binding capacity between lead ions and carbon dots.The degree of fluorescence intensity recovery showed a good linear relationship with the progestin concentration within the range of 9.90×10^(-7)-1.07×10^(-5)mol/L.The linear equation was I/I_(0)=5.83×10^(4)c_(progestin)+1.02 with a correlation coefficient of 0.9908.The detection limit was 3.7×10^(-7)mol/L.This method was applied to the detection of progestin concentration in injection solution, and the concentration of progestin ranged from 93.2% to 112%.
