LncRNA NEAT1通过调控miR-124-3p对食管鳞状细胞癌细胞迁移、侵袭的影响 被引量:3

摘要 目的 探讨长链非编码RNA核富集转录本1(LncRNA NEAT1)/微小RNA-124-3p(miR-124-3p)对食管鳞状细胞癌(ESCC)细胞迁移、侵袭的影响。方法 采用实时荧光定量PCR检测人ESCC细胞Eca109及正常人食管上皮细胞(HEEC)中LncRNA NEAT1及miR-124-3p表达水平;采用双荧光素酶报告基因实验验证LncRNA NEAT1与miR-124-3p的靶向关系;利用细胞转染技术分别将miR-124-3p抑制物、miR-124-3p模拟物、LncRNA NEAT1小干扰RNA(si-NEAT1)转染于Eca109细胞,采用Transwell实验检测Eca109细胞迁移、侵袭能力。结果 与HEEC细胞相比,Eca109细胞中LncRNA NEAT1表达水平明显升高(P<0.05),而miR-124-3p表达水平明显降低(P<0.05)。miR-124-3p和LncRNA NEAT1存在靶向关系,且LncRNA NEAT1可靶向负调控miR-124-3p表达水平。下调LncRNA NEAT1表达水平或过表达miR-124-3p均能抑制Eca109细胞迁移、侵袭能力,抑制miR-124-3p表达逆转了下调LncRNA NEAT1表达水平对Eca109细胞迁移、侵袭的作用。结论 下调LncRNA NEAT1表达水平可能通过靶向激活miR-124-3p表达来抑制ESCC细胞的迁移、侵袭能力。 Objective To investigate the effects of long non-coding RNA nuclear-enriched abundant transcript 1(LncRNA NEAT1) on the migration and invasion of esophageal squamous cell carcinoma cells(ESCC) by regulating microRNA-124-3 p(miR-124-3 p).Methods Real-time fluorescent quantitative PCR was used to detect the expression levels of LncRNA NEAT1 and miR-124-3 p in human ESCC cells Eca109 and normal human esophageal epithelial cells(HEEC);dual luciferase reporter gene assay was used to verify the targeting relationship between LncRNA NEAT1 and miR-124-3 p;cell transfection technology was used to transfect miR-124-3 p inhibitor, miR-124-3 p mimics, and LncRNA NEAT1 small interering RNA into Eca109 cells, and the Transwell test was used to detect the cell migration and invasion ability.Results Compared with HEEC cells, the expression level of LncRNA NEAT1 in Eca109 cells significantly increased(P<0.05),while the expression level of miR-124-3 p in Eca109 cells significantly decreased(P<0.05).There was a targeting relationship between miR-124-3 p and LncRNA NEAT1,and LncRNA NEAT1 could target and negatively regulate the expression level of miR-124-3 p.Down-regulation the expression level of LncRNA NEAT1 or over-expression of miR-124-3 p could inhibit the migration and the invasion of Eca109 cells, and inhibition the expression level of miR-124-3 p reversed the effect of down-regulation the expression level of LncRNA NEAT1 on the migration and invasion of Eca109 cells.Conclusion Down-regulation the expression level of LncRNA NEAT1 may inhibit the invasion and the migration of ESCC cells through targeting activation of miR-124-3 p expression.
