基于NLRP3炎症小体途径探究脊髓康促进脊髓损伤大鼠修复的机制 被引量:9

Mechanism of Jisui Kang in promoting the repair of spinal cord injury rats based on the NLRP3 inflammasome pathway

摘要 目的:基于NLRP3炎症小体途径探究脊髓康对脊髓损伤大鼠炎症反应的影响,并揭示其改善脊髓损伤后运动功能的部分机制。方法:采用改良Allen’s法制备SD大鼠脊髓损伤模型,分为模型组、强的松组、脊髓康组、假手术组。使用BBB评分和斜板实验评价大鼠的运动功能,提取血清后通过ELISA法检测血清中IL-1β、IL-18、IL-6、TNF-α的表达,Western Blot检测脊髓损伤区域NLRP3、ASC、NF-κB、Caspase-1表达,qRT-PCR检测脊髓损伤区域NLRP3、ASC、Caspase-1表达。结果:与假手术组同期比较,模型组大鼠斜板实验角度和BBB评分均降低,模型组第7天时IL-18、IL-6、TNF-α含量显著升高,ASC蛋白及mRNA表达显著升高,第28天时NLRP3、Caspase-1蛋白及mRNA表达升高(P<0.05,P<0.01);与模型组比较,脊髓康组在第21、28天斜板实验角度和BBB评分显著增加,在第7天时IL-18、IL-6、TNF-α含量显著降低,ASC蛋白及mRNA表达显著降低,第28天时NLRP3蛋白及mRNA表达显著降低(P<0.01)。结论:脊髓康可抑制NLRP3炎症小体途径,减轻脊髓损伤后炎症反应,从而改善脊髓损伤大鼠运动功能。 Objective: Based on the NLRP3 inflammasome pathway, to explore the effects of Jisui Kang on the inflammatory response in rats with spinal cord injury, and to reveal some of its mechanisms of improving motor function after spinal cord injury. Methods: The SD rat models of spinal cord injury were prepared by the modified Allen’s method, and they were divided into model group, prednisone group, Jisui Kang group and sham operation group. The motor function of rats was evaluated by BBB score and the oblique plate test. After serum was extracted, the expressions of IL-1β, IL-18, IL-6 and TNF-α in serum were detected by ELISA. Western Blot was used to detect NLRP3, ASC, NF-κB, Caspase-1 expression in the spinal cord injury area, qRT-PCR was used to detect of NLRP3, ASC, Caspase-1 expression in spinal cord injury area. Results: Compared with the sham operation group at the same time point, the model group had significantly reduced the angle of the oblique plate test and the BBB score at all time points, higher expression of IL-18, IL-6, TNF-α, higher expression of ASC protein and mRNA at the 7th day, and higher expression of NLRP3, Caspase-1 protein and mRNA at the 28th day(P<0.05, P<0.01). Compared with the model group,the angle of the oblique plate test and the BBB score significantly increased at the 21st and 28th day in JSK group;the expression of IL-18, IL-6, TNF-α significantly decreased at the 7th day, the expression of ASC protein and mRNA significantly decreased at the same time, and the expression of NLRP3 protein and mRNA significantly decreased at the 28th day(P<0.01).Conclusion: Jisui Kang can inhibit the NLRP3 inflammasome pathway and reduce the inflammatory response after spinal cord injury, thereby improving the motor function of rats with spinal cord injury.
