摘要
目的:探讨清瘟败毒饮对病毒性肺炎大鼠的作用和机制。方法:将18只Wistar大鼠随机分为空白对照组、病毒模型组、截断治疗组,每组6只。病毒模型组、截断治疗组予FM1(0.05 mL,LD)流感病毒液滴鼻,空白对照组予PBS液滴鼻。造模第4天每组各取3只大鼠,运用ELISA测定血清肿瘤坏死因子α(TNF-α)、干扰素γ(IFN-γ)、白介素(IL)-6、IL-10表达水平,用实时荧光定量PCR(RT-qPCR)检测肺组织T-Bet mRNA、GATA-3 mRNA表达情况。同时从造模第4天开始,截断治疗组给予清瘟败毒饮灌胃治疗4 d,空白对照组和病毒模型组给予同体积的0.9%氯化钠溶液灌胃。在第7天取剩余大鼠运用ELISA、RT-qPCR检测相关指标。结果:在流感病毒感染第4天,与空白对照组比较,病毒模型组IFN-γ、TNF-α、T-Bet mRNA显著升高(P<0.01)。流感病毒感染第7天,与空白对照组比较,病毒模型组和截断治疗组IFN-γ、TNF-α、T-Bet mRNA仍显著升高(P<0.05),病毒模型组IL-6、IL-10、GATA3 mRNA显著升高(P<0.01);与病毒模型组比较,截断治疗组TNF-α、IFN-γ、IL-6、IL-10,T-Bet、GATA3 mRNA显著降低(P<0.05,P<0.01)。结论:清瘟败毒饮能改善流感病毒感染大鼠肺组织病理状态,其机制可能与清瘟败毒饮调节Th1/Th2平衡有关。
Objective: To investigate the effect and mechanism of Qingwen Baidu Decoction on viral pneumonia in rats. Methods: Eighteen Wistar rats were randomly divided into blank control group (BC), severance treat group (ST) and virus model group (VM), 6 rats in each group. FM1 (0.05 mL, LD) influenza virus solution was given as nasal drops to the severance treat group and the virus model group, and PBS solution was given as nasal drops to the blank control group. On the 4th day of modeling, three rats were taken from each group, and the expression levels of serum TNF-α, IFN-γ, IL-6 and IL-10 were measured by ELISA, and the expression of T-Bet mRNA and GATA-3 mRNA in lung tissues were measured by realtime quantitative polymerase chain reaction (RT-qPCR). Meanwhile, from the 4th day of modeling, the severance treat group was given Qingwen Baidu Decoction by gavage for 4 days, and the blank control group and the virus model group were given the same volume of normal saline by gavage. The remaining rats were tested by ELISA and RT-qPCR on day 7. Results: On the 4th day of modeling, compared with the blank control group, IFN-γ, TNF-α and T-Bet mRNA in the viral model group were significantly increased (P<0.01). On the 7th day, compared with the blank control group, IFN-γ, TNF-αand T-Bet mRNA in the viral model group and the severance treat group were still increased (P<0.05 ), IL-6, IL-10 and GATA3 mRNA were significantly increased in the viral model group (P<0.01 ). Compared with the virus model group, TNF-α, IFN-γ, IL-6, IL-10, T-Bet mRNA,GATA3 mRNA in the severance treat group significantly decreased (P<0.05, P<0.01). Conclusion: Qingwen Baidu Decoction can ameliorate the pathological state of lung tissue in rats infected with influenza virus, and its mechanism may be related to the regulation of Th1/Th2 balance by Qingwen Baidu Decoction.
