CD8^(+)PD-1^(+)T细胞在胃癌患者外周血中的表型、功能及与临床分期的关系

Phenotype,Function and Clinical Stage of CD8^(+)PD-1^(+)T Cellsin Peripheral Blood of Patients with Gastric Cancer

目的探讨胃癌患者外周血中CD8^(+)程序性细胞死亡受体1(PD-1)+T细胞的表型、功能、分期间的关系。方法选择于2020年1月至2021年4月在我院就诊的42例胃癌患者为研究组,选择同一时期的42例健康受试者为对照组,对比血样的检测的检测结果,并分析与临床分期间的相关性。结果研究组CD8、CD8^(+)PD-1^(+)T细胞比例较高(P<0.05);初始型CD8^(+)PD-1^(+)T细胞占比低于CD8^(+)PD-1-T细胞占比,中心记忆型CD8^(+)PD-1^(+)T细胞比例高于CD8^(+)PD-1-T细胞比例,效应记忆型CD8^(+)PD-1^(+)T细胞比例高于CD8^(+)PD-1-T细胞比例,终末分化效应记忆型CD8^(+)PD-1^(+)T细胞比例低于CD8^(+)PD-1-T细胞比例(P<0.05)。晚期胃癌患者外周血中CD8^(+)PD-1^(+)T细胞比例高于早期,早晚期对比差异有统计学意义(P<0.05);结果显示,联合细胞中抗人干扰素-γ(IFN-γ)以及肿瘤坏死因子α(TNF-α)表达水平均低下(P<0.05)。结论CD8^(+)PD-1^(+)T细胞在胃癌中的表型、功能出现改变,与病情进展关系密切,为之后相关研究提供数据参考。

Objective To explore the relationship between phenotype,function and stage of CD8^(+)programmed cell death receptor 1(PD-1)+T cells in peripheral blood of patients with gastric cancer.Methods 42 patients with gastric cancer who visited our hospital from January,2020 to April,2021 were selected as the study group,while 42 healthy subjects during the same period were selected as the control group.The results of blood samples were compared and the correlation with clinical periods was analyzed.Results The proportion of CD8 and CD8^(+)PD-1^(+)T cells was higher in the study group(P<0.05),while the proportion of naive CD8^(+)PD-1^(+)T cells was lower than that of CD8^(+)PD-1-T cells,and the proportion of central memory type CD8^(+)PD-1^(+)T cells was higher than that of CD8^(+)PD-1-T cells.The proportion of effector memory type CD8^(+)PD-1^(+)T cells was higher than that of CD8^(+)PD-1-T cells,and the proportion of memory type CD8^(+)PD-1^(+)T cells with terminal differentiation effect was lower than that of CD8^(+)PD-1-T cells(P<0.05).The proportion of CD8^(+)PD-1^(+)T cells in peripheral blood of patients with advanced gastric cancer was higher than that in the early stage(P<0.05),and anti-human interferon-γ(IFN-γ)and tumor necrosis factorα(TNF-α)showed low expression levels(P<0.05).Conclusion The phenotype and function of CD8^(+)PD-1^(+)T cells in gastric cancer have changed compared with normal condition,which is closely related to the progress of the disease,and provides a reference for related research in the future.
