摘要
目的评价HPLC方法的可行性,为临床实验室不同检测系统检测血清中维生素A和维生素E结果的一致性提供参考。方法通过高效液相色谱(HPLC)与液相色谱-串联质谱(LC-MS/MS)检测人血清中维生素A/E的浓度的方法,进行比对。结果高效液相色谱法的方法学评价中,维生素A和维生素E的线性相关系数r>0.99,维生素A批内和批间精密度分别为0.70%~4.20%和0.10%~3.00%、维生素E批内和批间精密度分别为0.80%~6.40%和1.40%~3.10%,维生素A准确度为104.00%~111.25%、维生素E准确度为97.16%~106.12%,符合方法学要求。同时使用高效液相色谱和液相色谱-串联质谱检测191例临床样本中的维生素A和维生素E,得到结果进行Bland-Altman分析,维生素A和维生素E均满足95%的点都落在一致性界限内。结论两种方法检测临床样本中的维生素A和维生素E的结果基本没有差异。
Objective The feasibility of the HPLC method was evaluated by comparing high performance liquid chromatography(HPLC)with liquid chromatography-tandem mass spectrometry(LC-MS/MS)to detect the concentration of vitamin A/E in human serum.This experiment provided a reference for the consistency of concentrations of vitamin A and vitamin E in serum from different detection systems in different clinical laboratories.In the methodological evaluation of high performance liquid chromatography,the linear correlation coefficients of vitamin A and vitamin E were>0.99,the precision of vitamin A in and between batches were 0.70%-4.20%and 0.10%-3.00%and the precision of vitamin E in and between batches were 0.80%-6.40%and 1.40%-3.10%,while the accuracy of vitamin A was 104.00%-111.25%and the accuracy of vitamin E was 97.16%-106.12%.In summary,all results could meet the methodological requirements.At the same time,both high performance liquid chromatography and high performance liquid chromatography-tandem mass spectrometry were used to detect vitamin A and vitamin E in 191 clinical samples,and the results were obtained by Bland-Altman analysis.Both vitamin A and vitamin E meet the requirement that 95%of the points were within the consistency limit,indicating that there was no difference in the detection of vitamin A and vitamin E in clinical samples between the two methods.
