基于UPLC-ESI-HRMS^(n)技术的珍珠荚蒾果实抗肿瘤活性提取物的化学成分分析 被引量:1

摘要 为探究珍珠荚蒾果实的抗肿瘤活性及其组成成分,本文采用系统溶剂提取法制备了珍珠荚蒾果实石油醚、二氯甲烷、乙酸乙酯、正丁醇、水等5种提取物,并通过CCK8法评价这些提取物对人肝癌细胞(HepG2)、人宫颈癌细胞(Hela)、人非小细胞肺癌细胞(A549)、人乳腺癌细胞(MCF-7)的抑制作用.结果显示,珍珠荚蒾果实5种提取物对4种肿瘤细胞均有一定的抑制作用,以二氯甲烷提取物对Hela细胞的抑制作用最好,IC_(50)达到了116.8±5.35μg/mL.同时,综合UPLC-ESI-HRMS^(n)的分析结果和文献报道的数据从珍珠荚蒾果实二氯甲烷提取物种推断鉴定出39种化学成分,包括12种有机酸类化合物,10种萜类化合物,5种苯丙素类化合物,4种黄酮类化合物,3种香豆素类化合物,5种其他类化合物.结合文献对这些成分药理作用的报道,珍珠荚蒾果实中含有的三萜、黄酮、苯丙素等类型的成分可能是其发挥抗肿瘤作用的物质基础. To study the anti-tumor activities and components of Viburnum foetidum var.Ceanothoides fruits(V.Ceanothoides fruits),petroleum ether extracts,dichloromethane extracts,ethyl acetate extracts,n-butanol extracts and water extracts of V.Ceanothoides fruits were prepared by systematic solvent extraction.Subsquently,the inhibitory effects of the five extracts on human hepatoma cells(HepG2),human cervical cancer cells(Hela),human non-small-cell lung carcinoma cells(A549)and human breast cancer cells(MCF-7)were evaluated by CCK8.The results showed that the five extracts of V.Ceanothoides fruits could inhibit the growth of 4 kinds of tumor cells(HepG2,Hela,A549 and MCF-7),and the dichloromethane extracts had the most potential to inhibit the growth of Hela cells with IC_(50) of 116.8±5.35μg/mL.Based on the results of UPLC-ESI-HRMS^(n) and the data reported in the literature,39 chemical constituents,including 12 organic acids,10 terpenes,5 phenylpropanoids,4 flavonoids,3 coumarins and 5 other compounds,were identified from the dichloromethane extracts of V.Ceanothoides fruits.Triterpenoids,flavonoids and phenylpropanoids in Viburnum foetidum fruits may be the material basis of its anti-tumor effect according to the literature.
