摘要
从新疆阜康市和库尔勒市的苹果及香梨根际土壤中分离到一种线虫,通过形态特征观察、形态测计,并提取DNA,扩增其18S rDNA、28S rDNA D2-D3和ITS区基因片段,利用NJ法构建系统进化树,进行种类鉴定。结果表明,供试线虫群体形态学特征与畸形轮线虫山东种群、荷兰种群测计值基本一致,构建的18S rDNA区、28S rDNA D2-D3区和ITS区序列系统进化树与NCBI上记录的畸形轮线虫聚为同一支。结合形态学特征与分子生物学特征,将该新疆线虫种群鉴定为畸形轮线虫(Criconemoides informis),也是该线虫在新疆果树上的首次报道。
A population of nematode was isolated from the root soil of Malus pumila and Pyrus sinkiangensi in Fukang and Korla,Xinjiang,and identified by morphological feature observation and morphometry.The DNA was extracted and its 18S rDNA,28S rDNA D2-D3 and ITS region gene fragments were amplified,and phylogenetic tree was constructed by using the NJ method for species identification.The results showed that the morphological feature of the nematode population was consistent with the morphometric values of the Shandong and Dutch populations of the nematode,the constructed phylogenetic trees of the 18S rDNA,28S rDNA D2-D3 and ITS regions were all clustered together with the Criconemoides informis recorded on NCBI.Combining the morphological and molecular biological characters,the Xinjiang nematode population was identified as Criconemoides informis,which was the new record of this species of nematode on fruit trees in Xinjiang.
