克里本类芽孢杆菌TRCC 82001抑菌活性分析及发酵条件优化 被引量:1

摘要 分析克里本类芽孢杆菌TRCC 82001的抑菌活性和防病效果,优化其发酵条件以提升发酵液抑菌效果。通过平板对峙试验测定拮抗菌的抑菌活性,利用不同检测平板分析其产真菌细胞壁裂解酶特性,以单因素试验筛选其最优发酵培养基和最优发酵条件,最后通过离体枝条防治试验研究其防病能力。结果表明,克里本类芽孢杆菌TRCC 82001对金黄壳囊孢菌等多种植物病原真菌均有较好的抑菌活性,抑菌带宽度范围在10.00~13.33 mm;能产生蛋白酶、β-1,3-葡聚糖酶和纤维素酶;最优发酵培养基为马铃薯葡萄糖肉汤培养基;最佳发酵条件组合为时间3 d、温度30℃、pH 6.0、转速180 r/min、装液量20%和接种量1%;拮抗菌在离体枝条上对杨树腐烂病的抑制效果达到100%。该研究结果为该菌株及其抑菌物质在防治植物病害方面的应用奠定了基础。 The objectives of this study were to analyze the antifungal activity and control effect of Paenibacillus kribbensis TRCC 82001,and optimize the fermentation conditions to enhance the antifungal activity of the fermentation broth.Antifungal activity of TRCC 82001 was tested by using the dual culture assay.The activities of different cell wall lyase were detected by different test medium plates.The fermentation medium and conditions were optimized according to single factor test.Antifungal activity was tested on detached twigs of Populus×euramericana.The results revealed that the strain TRCC 82001 had good antifungal activity against C.chrysosperma and other plant pathogenic fungi,the width of the inhibition zones were between 10.00-13.33 mm.It exhibited cell wall degrading traits by producing three lytic enzymes(protease,β-1,3-glucanase,and cellulose).The optimal fermentation conditions were as follows:medium:potato dextrose broth,time of fermentation:3 days,temperature:30℃,pH:6.0,rotate speed:180 r/min,bottling quantity:20%,and inoculum size:1%.The control effect of TRCC 82001 against C.chrysosperma was up to 100%on detached twigs.These results laid a foundation for the application of this strain and its antifungal substances in plant disease control.
