TRPV4和TGF-β1在口腔鳞状细胞癌中的表达及临床意义 被引量:3

The expression and significance of TRPV4 and TGF-β1 in OSCC

摘要 目的:研究口腔鳞状细胞癌(OSCC)组织中瞬时性受体电位通道香草酸受体4(TRPV4)和转化生长因子β1(TGF-β1)的表达及临床意义。方法:采用免疫组化EnVision法(ELPS)检测TRPV4和TGF-β1在103例OSCC组织及66例癌旁正常组织中的表达,分析与OSCC临床特点及预后之间的关系。结果:OSCC组织中TRPV4的阳性率69.90%,TGF-β1的阳性率74.76%,均明显高于癌旁正常组织中的表达(24.24%和19.70%)(P<0.05)。TRPV4和TGF-β1诊断OSCC的ROC曲线下面积(AUC)分别为0.842和0.857。TRPV4的表达与TNM分期和分化程度有关(P<0.05),与年龄、性别、淋巴结转移、浸润深浅无关(P>0.05);TGF-β1的表达与浸润深度和淋巴结转移有关(P<0.05)。OSCC中TRPV4、TGF-β1的表达呈正相关(r=0.211、P=0.033)。Kaplan-Meier生存分析显示,TRPV4和TGF-β1高表达与患者预后有关(P=0.0193,P<0.05)。结论:TRPV4和TGF-β1在组织中高表达,且与患者的不良预后密切相关。 Objective:To investigate the expression of TRPV4 and TGF-β1 in oral squamous cell carcinoma(OSCC)and its clinical significance.Methods:Immunohistochemistry EnVision method(ELPS)was used to detect the expression of TRPV4 and TGF-β1 levels in 103 OSCC specimens and 66 paracancerous specimens(PC)respectively.Results:The positive rate of TRPV4 and TGF-β1 in OSCC was higher than that in PC(69.90%and 74.76%vs 24.24%and 19.70%,P<0.05).The AUC area under the ROC curve(AUC)in the diagnosis of OSCC by TRPV4 and TGF-β1 was 0.842 and 0.857 respectively.The expression of TRPV4 was associated with the differentiation degree and TNM stage of OSCC(P<0.05),rather than age,sex,presence of axillary lymph node metastasis and the invasion depth measurement(P>0.05);the expression of TGF-β1 was associated with lymph node metastasis and the invasion depth measurement(P<0.05).The expression level displayed a positive correlation between TRPV4 and TGF-β1 in OSCC(r=0.211、P=0.033).The Kaplan-Meier Survival Analysis showed that the high expression of TRPV4 and TGF-β1 was associated with poor prognosis in OSCC patients.Conclusion:The expression of TRPV4 and TGF-β1 in OSCC is upregulated and is correlated with tumor progression and poor prognosis of the OSCC patients.
