摘要 为了建立一种不同加工工艺果汁DNA提取方法的通用方法,比较了改良CTAB法、试剂盒法和改良试剂盒法3种方法提取鲜榨果汁、浓缩果汁、非浓缩果汁、含果肉饮料4种类型果汁中DNA。结果表明:3种方法提取的DNA纯度相似,但提取的复原果汁DNA纯度高于2.0,可能是DNA降解造成;质量浓度由高到低分别为改良试剂盒法、改良CTAB法、试剂盒法,改良试剂盒法提取的DNA可以用作后续的一系列PCR反应。通过比较果汁DNA提取的3种方法,针对鲜榨果汁、浓缩果汁、非浓缩果汁、含果肉饮料中DNA,改良试剂盒法可以作为果汁中DNA提取的通用方法。 In order to establish a general method of extracting DNA from juice with different processing,DNA was extracted from fresh juice,concentrated juice,non-concentrated juice and pulp beverage by three methods including improved CTAB method,kit method and improved kit method.The purity of DNA extracted by the three methods was similar,but the purity of DNA extracted from the recovered juice was higher than 2.0,which might be caused by DNA degradation.The mass concentration from high to low were modified kit method,modified CTAB method and kit method respectively.The DNA extracted by the modified kit method could be used for a series of subsequent PCR reactions.By comparing the three methods of DNA extraction from fruit juice,it was pointed out that the improved kit method can be used as a general method for DNA extraction from fresh fruit juice,concentrated fruit juice,non-concentrated fruit juice and pulp drinks.
