摘要
桃子采后软腐病主要是由匍枝根霉(Rhizopus stolonifer)引起的真菌性病害。本研究通过体外抑菌试验发现,杰米拉类芽孢杆菌(Paenibacillus jamilae)W51(以下简称W51)直接抑制R.stolonifer孢子萌发及菌体生长,抑制率达到75.00%。用浓度为1×10^(8)CFU/mL的W51接种处理桃子,结果表明,W51处理组∶对照组的病斑直径比为1∶4.14,显著降低了果实的发病率和病斑直径。转录组测定结果显示,在贮藏第2天,W51处理诱导了编码ROS代谢相关PXMP2蛋白等差异基因,还引发了POD、SOD以及CHI相关基因的上调表达。qRT-PCR结果也表明,W51处理组的POD抗病相关基因和编码PXMP2蛋白相关差异基因的上调明显高于对照组。因此,生防菌W51处理能够显著促进桃果实对软腐病的抗性,提高其保鲜效果。
Peach postharvest soft rot is a fungal disease caused by Rhizopus stolonifer.In this study,we found that Paenibacillus jamilae W51(hereinafter referred to as W51) directly inhibited R.stolonifer.The inhibition rate of spore germination and mycelium growth reached 75.00%.W51(1×10^(8)CFU/m L) was inoculated to treat peaches.The results showed that the lesion diameter in W51 treatment group was 1∶4.14 compared with control group,significantly reducing the disease incidence and lesion diameter of fruits.Transcriptome analysis showed that W51treatment induced the up-regulated expression of differential genes encoding PXMP2 protein and other genes for ROS metabolism,as well as up-regulated expression of POD,SOD and CHI related genes on the second day of storage.The qRT-PCR results showed that on the second day of storage,the upregulation of POD disease resistance related genes and PXMP2 protein related differential genes in W51 treatment group were significantly higher than those in control group.Therefore,W51 treatment could significantly promote the resistance of peach fruit to soft rot and improve its fresh-keeping effect.
