肺炎患儿miR-147a、miR-217的表达及其与肺功能、炎症因子的相关性 被引量:3

Expression levels of miR-147a and miR-217 and their correlations with pulmonary function parameters and inflammatory factors in children with pneumonia

摘要 目的研究肺炎儿童血清miR-147a、miR-217的表达水平及与肺功能、炎症因子的关系。方法选取2019年1月至2021年12月于青岛市市立医院接受治疗的100例肺炎患儿为肺炎组,同期参加健康体检的41例同龄儿童为对照组。检测并比较两组miR-147a、miR-217表达水平,肺功能指标[呼气流量(PEF),第一秒用力呼气容积(FEV1),用力肺活量(FVC)、FEV1/FVC,25%、50%、75%肺活量的呼气流速V_(25)、V_(50)、V_(75)],炎症因子[白细胞介素-6(IL-6),IL-1β,肿瘤坏死因子α(TNF-α)];Pearson分析相关性。结果肺炎组血清miR-147a、miR-217表达量均低于对照组,差异有统计学意义(t值分别为4.302、5.322,P<0.05);肺炎组儿童PEF、FEV1、FEV1/FVC、V_(25)、V_(50)、V_(75)水平均低于对照组,差异有统计学意义(t值分别为5.921、6.035、8.392、5.772、7.274、9.926,P<0.05);肺炎组IL-6、IL-1β、TNF-α水平均高于对照组,差异有统计学意义(t值分别为28.372、9.372、11.285,P<0.05);miR-147a、miR-217表达与肺功能指标呈正相关(r值介于0.654~0.772之间,P<0.05),与炎症因子呈负相关(r值介于-0.811~-0.784之间,P<0.05),差异均有统计学意义。结论儿童肺炎血清中miR-147a和miR-127表达下调,且与肺功能指标呈正相关,与炎症因子呈负相关,miR-147a和miR-127可能是潜在的肺炎诊疗靶点。 Objective To explore relationship between expression levels of serum microRNA-147a(miR-147a)and microRNA-217(miR-217)and pulmonary function parameters and inflammatory factors in children with pneumonia.Methods A total of 100children with pneumonia who were treated in Qingdao Municipal Hospital from January 2019to December 2021were selected as pneumonia group,and 41children of the same age who received physical examination in our hospital in the same period were selected as control group.The expression levels of miR-147,miR-217,pulmonary function parameters[peak expiratory flow(PEF),forced expiratory volume in the first second(FEV1),forced vital capacity(FVC),FEV1/FVC,and maxinal expiratory flow in 25%,50%and 75%vital capacity(V_(25),V_(50)and V_(75))]and inflammatory cytokines(IL-6,IL-1βand TNF-α)of the children in the two groups were detected and compared.Pearson correlation analysis was used to analyze the correlations.Results The expression levels of miR-147aand miR-127of the children in the pneumonia group were lower than those in the control group,and the differences were statistically significant(t=4.302and 5.322respectively,both P<0.05).The levels of PEF,FEV1,FEV1/FVC,V_(25),V_(50)and V_(75)of the children in the pneumonia group were lower than those in the control group,and the differences were statistically significant(t=5.921,6.035,8.392,5.772,7.274and 9.926respectively,all P<0.05).The serum levels of IL-6,IL-1βand TNF-αof the children in the pneumonia group were higher than those in the control group,the differences were statistically significant(t=28.372,9.372and 11.285respectively,all P<0.05).MiR-147aand miR-217were positively correlated with pulmonary function parameters(r value ranged from 0.654to 0.772,all P<0.05),and they were negatively correlated with inflammatory factors(r value ranged from-0.811to-0.784,all P<0.05),and the differences were statistically significant.Conclusion The expressions of miR-147a and miR-127in the serum of children with pneumonia are down-regulated,and they are positively correlated with pulmonary function parameters and negatively correlated with inflammatory factors.It is preliminarily explored that miR-147aand miR-127may be potential diagnostic and therapeutic targets for pneumonia.
