摘要
去乙酰化酶SIRT1在衰老中起着关键作用,可通过p53、哺乳动物雷帕霉素靶蛋白(mTOR)、核因子-κB(NF-κB)、AMP活化蛋白激酶(AMPK)、过氧化物酶体增殖物激活受体γ共激活因子1α(PGC1α)信号转导途径发挥神经保护、抗氧化应激、抑制炎症反应和调控自噬等作用干预阿尔茨海默病(AD)进展。本文旨在回顾SIRT1抗衰老机制与AD之间的关系,探讨基于调节SIRT1活性干预AD的防治策略。
The deacetylase, inflammatory factor silencing information regulating protein1(SIRT1) plays a key role in aging.It exerts neuroprotective, antioxidant stress, inhibition of inflammatory response and regulation of autophagy to intervene in the progress of Alzheimer′s disease(AD) through some signaling pathways, such as p53,mTOR,NF-κB,AMPK and PGC1α.This article reviews the relationship between SIRT1 anti-aging mechanisms and AD,and discusses the prevention and treatment strategies of AD based on regulating SIRT1 activity.
