摘要
目的:观察亲环素A(CypA)、CD147及基质金属蛋白酶2(MMP-2)在人慢性牙周炎患者牙龈组织中CD68^(+)细胞(单核细胞/巨噬细胞)的表达情况,探讨CD68^(+)细胞中CypA、CD147及MMP-2在不同病变程度牙周炎牙龈组织中的表达及作用机制。方法:将60例受试者分为4组,每组15例,分别为:健康组及慢性牙周炎(轻、中、重)组。免疫荧光双染色(DIF)后荧光显微镜下分别观察牙龈组织中CD68-CypA、CD68-CD147及CD68-MMP-2双阳性细胞表达情况并计算单位面积双阳性细胞密度。结果:(1)各组牙龈组织中CD68-CypA、CD68-CD147及CD68-MMP-2双阳性细胞密度差异显著(P<0.01);(2)各组牙龈组织中CD68-CypA双阳性细胞(r=0.959,P=0.000)、CD68-CD147双阳性细胞(r=0.948,P=0.000)及CD68-MMP-2双阳性细胞密度(r=0.945,P=0.000)与牙周炎的严重程度呈正相关;(3)各组牙龈组织中CD68-CypA与CD68-MMP-2双阳性表达正向相关(r=0.933,P=0.000);CD68-CD147与CD68-MMP-2双阳性表达水平正相关(r=0.954,P=0.000)。结论:牙龈组织中CD68-CypA、CD68-CD147及CD68-MMP2双阳性细胞密度随牙周病程加重而增多,且CD68-CypA和CD68-CD147双阳性细胞的密度与CD68-MMP-2双阳性细胞密度呈正相关。
AIM:To explore the function and mechanism of cyclophilinA(CypA),CD147 and matrix metalloproteinase-2(MMP-2)in gingival tissues with different degrees of periodontitis.METHODS:A total of 60 volunteers from the outpatient department were divided into 4 groups such as healthy group and different degrees of periodontitis.The expression of CD68-CypA,CD68-CD147 and CD68-MMP-2 double positive cells in gingival tissue was observed by double immunofluorescence staining(DIF).The density of double positive cells per unit area was calculated and analyzed.RESULTS:(1)Significant difference in cell densities of CD68-CypA positive,CD68-CD147 positive and CD68-MMP-2 double positive cells was observed in gingival samples(P<0.01);(2)CD68-CypA double positive cells(r=0.959,P=0.000),CD68-CD147 double positive cells(r=0.948,P=0.000)and CD68-MMP-2 double positive cell density(r=0.945,P=0.000)were positively related to the severity of periodontitis in gingival tissues of each group;(3)The double positive expression of CD68-CypA was positively related to CD68-MMP-2 in gingival tissues of each group(r=0.933,P=0.000).A significant positive correlation was found between the double positive expression of CD68-CD147 and CD68-MMP-2(r=0.954,P=0.000).CONCLUSION:The cell densities of CD68-CypA,CD68-CD147 and CD68-MMP-2 double positive cells in gingival tissue were increased with the progression of periodontitis,and were positively correlated with the inflammation score.And the cell densities of CD68-CypA and CD68-CD147 double positive cells in gingival tissue were positively correlated with CD68-MMP-2 double positive cells.
