可溶性环氧化物水解酶在病理性心脏重构中的意义及研究进展

摘要 心血管疾病是严重危害人类健康的重要疾病,其发病率一直居高不下,是导致城乡居民死亡的首要原因。心力衰竭(简称心衰)是多种心血管疾病的最终临床转归,而心脏重构是心衰的重要病理基础,其特征为伴有胚胎基因再表达的病理性心肌细胞肥大和心肌细胞外基质过度纤维化或降解增加[1]。目前临床上缺乏对病理性心脏重构的特异性干预方法,因而对心衰的治疗仍面临未能从根本上阻断其发展进程的现状。 Soluble epoxide hydrolase(sEH)is a key enzyme involved in cytochrome P450-mediated metabolism of polyunsaturated fatty acids. This article presents a brief overview of the role of sEH in the development of pathological cardiac remodeling. We reviewed the role of sEH in hypertension,cardiomyopathy and ischemic heart diseases,which are the common causes of cardiac remodeling,and highlighted the multiple interrelated mechanisms involved in sEH-mediated cardiac remodeling such as mediation of oxidative and endoplasmic reticulum stress,promotion of inflammatory response,bi-directional regulation of autophagy,and induction of mitochondrial damage. This review also focuses on the development of sEH-targeting drugs and their clinical application prospects in the treatment of cardiac remodeling.
