摘要 目的 建立一种高效、准确的鉴别月经血与外周血的方法体系并探究该体系的应用能力。方法 设计miR-451a、miR-21-5p的引物、探针,建立并优化基于TaqMan技术的qPCR检测体系;对58份样本(20份月经血、20份外周血、6份唾液、6份精液、6份阴道分泌液)中miR-451a与miR-21-5p的表达情况进行检测,探索候选miRNA分子在5种体液中的表达规律;最终建立区分目标体液的鉴别方法并用模拟案件样本对其准确性进行验证。结果 本研究所建立体系及方法可实现血液与非血液、月经血与外周血100%准确区分。结论 本研究基于TaqMan qPCR技术成功建立了运用miR-451a/miR-21-5p表达量比值鉴别月经血与外周血的方法体系,该体系鉴别能力良好,有望应用于实际法医案件中。 Objective To establish an efficient and accurate system to distinguish menstrual blood from peripheral blood and explore the application value of this system.Method Firstly,the primers and probes targeting miR-451a and miR-21-5p respectively were designed,and a qPCR detection system based on the TaqMan technology was established and optimized.Secondly,we detected the expression levels of miR-451a and miR-21-5p in 58 samples(including 20menstrual blood,20 peripheral blood,6 saliva,6 semen,6 vaginal secretion samples).Finally,the identification method was established and its accuracy was then verified using mock case samples.Results The system established in this study could distinguish blood from non-blood and menstrual blood from peripheral blood with a accuracy of 100%.Conclusion In this study,based on the TaqMan qPCR technology,a method was successfully established to differentiate menstrual blood from peripheral blood by using the miR-451a and miR-21-5p expression ratio.This system has a good discrimination ability and is expected to be applied in practical forensic cases.
