摘要
目的:研究互叶白千层挥发油的化学成分组成及其抗肿瘤和抗虫生物活性。方法:采用气相色谱-质谱联用法分析互叶白千层挥发油的化学成分组成,采用CCK-8法检测挥发油及其主要成分对MDA-MB-231和B16细胞增殖的抑制作用,采用熏蒸、触杀和驱避活性测试评价挥发油及其主要成分对中药仓储害虫赤拟谷盗的抗虫作用。结果:互叶白千层挥发油的主要成分为(-)-萜烯-4-醇(36.02%)、γ-松油烯(21.42%)、2-蒈烯(10.56%)、萜品油烯(3.34%)和1,8-桉树脑(3.17%)等。互叶白千层挥发油及γ-松油烯对B16细胞的增殖具有显著的抑制作用,IC_(50)分别为(12.02±1.46)和(17.47±5.38)μg/ml。1,8-桉树脑、γ-松油烯和(-)-萜烯-4-醇对赤拟谷盗具有显著的熏蒸活性,LC_(50)均小于18 mg/L;γ-松油烯对赤拟谷盗具有显著的触杀活性,LD_(50)为17.67μg/头;挥发油对赤拟谷盗的驱避活性最为显著,ri值为0.6601。结论:互叶白千层挥发油及其活性成分具有抗肿瘤及抗虫作用。
Objective:To investigate the chemical constituents of the essential oil from Melaleuca alternifolia and its anti-tumor and anti-insect activities.Methods:The chemical constituents of essential oil from Melaleuca alternifolia were identified with gas chromatography-mass spectrometry.The inhibitory effect of the essential oil and its main components on the proliferation of MDA-MB-231 and B16 cells was detected by CCK-8 assay.Anti-insect activities of the essential oil and its main components against Tribolium castaneum were evaluated by fumigant,contact and repellent tests.Results:(-)-Terpinen-4-ol(36.02%),γ-terpinene(21.42%),2-carene(10.56%),isoterpinolene(3.34%)and 1,8-cineole(3.17%)were found to be major components of the essential oil from Melaleuca alternifolia.The essential oil andγ-terpinene significantly inhibited the proliferation of B16 cells,with the IC_(50)values of(12.02±1.46)and(17.47±5.38)μg/ml,respectively.1,8-Cineole,γ-terpinene and(-)-terpinen-4-ol exhibited significant fumigant activity against Tribolium castaneum,with LC50 values less than 18 mg/L.γ-Terpinene exerted strongest contact activity against Tribolium castaneum,with LD50 value of 17.67μg/adult.The essential oil showed marked repellent activity against Tribolium castaneum with ri value of 0.6601.Conclusion:The essential oil from Melaleuca alternifolia and its major constituents exhibited anti-tumor and anti-insect activities.
