芍药甘草汤相态拆分及镇痛抗炎活性评价 被引量:1

摘要 目的对芍药甘草汤进行相态拆分,并对其进行成分分布及镇痛抗炎药效学评价。方法采用离心及膜抽滤的方法进行芍药甘草汤不同相态的拆分;通过粒径分析和形态观察对拆分后的纳米相态进行确认;HPLC法测定芍药甘草汤5个指标性成分在不同相态中的分布;以醋酸致小鼠扭体和二甲苯致小鼠耳肿胀为研究模型,观察芍药甘草汤不同相态以及活性成分含量与镇痛抗炎效果的相关性。结果芍药甘草汤纳米相态的粒径约为250 nm,呈球形;芍药苷、芍药内酯苷在真溶液相态的分布>纳米相态>沉淀相态,而甘草酸、甘草苷、芹糖甘草苷在纳米相态的分布>真溶液相态>沉淀相态;芍药甘草汤纳米相态的抗二甲苯致小鼠耳肿胀和抑制醋酸致小鼠扭体的作用显著优于沉淀相态和真溶液相态(P<0.05),与全方的药效相当(P>0.05)。结论芍药甘草汤不同相态的活性成分存在空间分布差异,其镇痛抗炎作用与芍药甘草汤活性成分含量及其物理结构形式密切相关。芍药甘草汤纳米相态的镇痛抗炎效果显著优于其他相态,是其发挥镇痛抗炎作用的关键。 Objective To study the phase separation,composition distribution and therapeutic effect of shaoyao gancao decoction(SGD).Methods The phase separation of SGD was performed via centrifugation and membrane filtration.The obtained nano phase states were characterized by means of particle size analysis and morphological observation.The five components of SGD in different phases were determined by HPLC.The analgesic effect of different phases of SGD in acetic acid-induced writhing model mice and the anti-inflammatory effect of different phases of SGD in xylene-induced ear swelling model mice were investigated.Results The nano phase of SGD had a particle size of about 250 nm and the morphology was spherical in shape.Albiflorin and paeoniflorin were dominatingly distributed in the true solution phase,followed by the nano phase and precipitation phase while liquiritin apioside,liquiritin and glycyrrhizic acid were mostly distributed in the nano phase,followed by the true solution phase and precipitation phase.SGD was much more effective for xylene-induced ear swelling of mice and acetic acid-induced writhing of mice in the nano phase than in the precipitation phase and true solution phase(P<0.05).Conclusion The analgesic and anti-inflammatory effects of SGD are better in the nano phase than in other phases,indicating that the nano phase of SGD plays a pivotal role in the analgesic and anti-inflammatory effect of SGD.
