Characteristic Peptides of Periplaneta Americana

目的对美洲大蠊特征肽段进行研究。方法采用碱裂解法提取蛋白质,胰蛋白酶水解,离子阱静电轨道阱组合式高分辨质谱结合分析软件进行多肽特征离子对的寻找,最后采用三重四级杆质谱对特征性离子对进行验证和多反应监测模式研究。结果发现并确证了质荷比(m/z)510.8(双电荷)→369.0,m/z 510.8(双电荷)→239.9;质荷比(m/z)627.3(三电荷)→603.5,m/z 627.3(三电荷)→516.7离子对为美洲大蠊特征性离子对。而美洲大蠊的混伪品德国小蠊、杜比亚、土鳖虫均未出现这两对特征性离子对。结论发现并建立美洲大蠊多肽特征离子对鉴别方法,专属性强,对原料药的掺杂混伪品情况进行严格控制。

Objective To study the characteristic peptides of Periplaneta americana.Methods The main protein components of Periplaneta americana were extracted using a alkaline lysis method and diges-ted by trypsin.The obtained peptides were analyzed with ion trap high resolution mass spectrometry and the data was processed.The characteristic peptides of Periplaneta americana were screened out,the speci-ficity of which was verified with triple quadrupole mass spectrometry.Results The marker peptides-m/z 510.8(double-charged)→369.0,m/z 510.8(double-charged)→239.9 and m/z 627.3(three-charged)→603.5,m/z 627.3(three-charged)→516.7-of Periplaneta americana were found,which had never been discovered in Blattella Germanica,Blaptica Dubia or Eupolyphaga sinensis Walker.Conclusion The characteristic peptides can be used for specificity identification of Periplaneta americana from the adulter-ant,which is of great significance for its quality control.
