摘要
目的:通过鉴定27份番茄种质资源的亲缘关系,以提高番茄育种效率。方法:利用RAPD子标记技术和SPSS 16.0软件对番茄种质材料进行遗传多样性及亲缘关系聚类分析。结果:60条随机引物的有效率为63.3%,27份番茄种质材料PCR共扩增出196条带,多态性条带190条,多态性比率96.93%;当欧氏距离D值在17.5时,可将27份番茄分为4类。结论:RAPD分子标记技术可从分子视角给予番茄种质材料较全面、客观的评价与分析,为番茄优质品种的选育提供参考依据。
Objective:In order to determine more accurately the genetic relationship of 27 tomato germplasm resources and improve the tomato breeding efficiency.Methods:RAPD-PCR molecular marker technology and SPSS 16.0 software were used to analyze the genetic diversity and phylogenetic relationship of tomato germplasms.Results:The effective percentage of 60 random primers was 63.3%.A total of 196 bands,within 190 polymorphic bands,were amplified by PCR from 27 tomato germplasm materials and the polymorphism ratio was 96.93%.The genetic diversity analysis showed that 27 tomato germplasm resources could be divided into 4 groups at the genetic distance D value of 17.5.Conclusion:RAPD molecular marker technology can give a more comprehensive and objective evaluation and analysis of tomato germplasm materials from a molecular perspective,and provide a reference of parental pairing for breeding of high-quality tomato varieties.
