甘薯根际土壤微生物群落结构及多样性分析 被引量:13

摘要 【目的】通过探究不同品种甘薯根际土壤细菌和真菌群落结构及其分布规律,旨在探讨根际细菌和真菌对甘薯生长发育的影响。【方法】提取龙薯24、普薯32和福宁紫3号3个甘薯品种根际土壤微生物的总DNA,采用高通量测序技术对样品进行测序,分析研究细菌和真菌群落结构和多样性。【结果】3个品种土壤样品中共获得细菌OTUs 2978个,真菌OTUs 686个。物种分类显示细菌种类隶属于33门91纲222目364科630属1248种,真菌隶属于13门30纲61目108科163属213种,细菌群落的多样性和丰富度均高于真菌。不同品种甘薯根际土壤样品中细菌和真菌群落组成和结构均存在一定差异,细菌丰富度和多样性为:福宁紫3号>普薯32>龙薯24,真菌丰富度和多样性为普薯32>龙薯24>福宁紫3号。细菌群落中变形菌门(Proteobectria)、绿弯菌门(Chloroflexi)、放线菌门(Actinobacteria)和酸杆菌门(Acidobacteria)4个菌门相对丰度均大于10%,丰度之和达79.38%~82.21%,是绝对优势菌门。真菌群落中子囊菌门(Asccomycota)在龙薯24、普薯32和福宁紫3号样品中相对丰度分别高达81.4%、57.03%和72.24%,是最绝对优势菌门。【结论】不同品种甘薯根际土壤样品中的细菌和真菌群落结构均存在一定差异,变形菌门(Proteobectria)、绿弯菌门(Chloroflexi)、放线菌门(Actinobacteria)和酸杆菌门(Acidobacteria)4个菌门是细菌绝对优势菌门,子囊菌门(Asccomycota)是真菌绝对优势菌门。 【Objective】The presnet paper aimed to reveal the effects of rhizophere bacteria and fungi on growth of sweet potato,we analyzed bacteria and fungi community structures and genetic diversities in rhizosphere soil of three different sweet potato varieties.【Method】Rhizophere soil microbial total DNA of three sweet potato varieties(Longshu24,Pushu32,and Funingzi3)was extracted.High-throughput sequencing technologies was employed to analyze the soil bacterial and fungal community structure and diversity.【Result】2978 and 686 OTUs were obtained from soil bacteria and fungi.Based on the results of species classification,bacterial communities belonging to 33 phyla,91 classes,222 orders,364 families,630 genera and 1248 species,while fungal species belonged to 13 phyla,30 classes,61 orders,108 families,163 genera and 213 species.Bacterial communities were more diverse and had greater species richness than fungal community in rhizosphere soil.There were some differences in the structure and composition of bacterial and fungal community in rhizosphere soil samples of different sweet potato varieties.In terms of bacterial richness and diversity,Funingzi3>Pushu32>Longshu24,while in terms of fungal richness and diversity,Pushu32>Longshu24>Funingzi3.Among all the bacterial community,the dominant populations were Proteobectria,Chloroflexi,Actinobacteria and Acidobacteria,of which the relative abundance exceeding 10%,and sum of the four community relative abundance was 79.38%to 82.21%.The dominant fungal species was Asccomycota,and the relative abundance of Longshu24,Pushu32 and Funingzi3 reached 81.4%,57.03%and 72.24%.【Conclusion】There were some differences in the structure and composition of bacterial and fungal community in rhizosphere soil samples of different sweet potato varieties.Proteobectria,Chloroflexi,Actinobacteria and Acidobacteria were dominant bacterial species,and Asccomycota was the dominant fungal species.
