CELF1在女性PTC组织中的表达及其对细胞增殖和侵袭力的影响研究

摘要 目的:研究女性甲状腺乳头状癌(PTC)组织中RNA结合蛋白家族1(CELF1)的表达情况及CELF1的临床意义。方法:以福州市长乐区医院2021年1月-2022年6月女性PTC患者86例为研究对象,采用回顾性研究,根据免疫组织化学法检测CELF1蛋白在PTC组织与癌旁组织中的阳性表达率;通过小分子RNA建立CELF1低表达细胞系,并建立空白对照组、阴性对照组和试验组。测定CELF1 mRNA表达情况,分别采用CCK-法与Transwell小室实验检测三组细胞增殖和侵袭能力。结果:CELF1蛋白在PTC组织中的阳性表达率为75.58%(65/86),较癌旁组织的39.53%(34/86)高(χ^(2)=22.87,P<0.01)。试验组PTC细胞中CELF1 mRNA表达量为(1.06±0.09),较空白对照组的(2.21±0.16)和阴性对照组的(2.33±0.13)均下调(F=154.76,P<0.01)。经细胞培养1、2 d后,相比空白对照组和阴性对照组,试验组细胞增殖能力明显减弱(F=47.95、56.72,P<0.01)。相比空白对照组(122.86±15.34)个和阴性对照组(120.57±13.54)个,试验组细胞侵袭数量(84.13±10.04)个较少(F=60.24,P<0.01)。结论:PTC组织中CELF1具有高表达的特点,而通过特异性降低CELF1 mRNA在PTC细胞中的表达量可起到抑制细胞增殖和降低细胞侵袭的作用。 Objective:To study the expression of RNA binding protein family 1 (CELF1) in female papillary thyroid carcinoma (PTC) and CELF1’s clinical significance.Method:A total of 86 female PTC patients in Fuzhou City Changle District Hospital from January 2021 to June 2022 were taken as the research objects,a retrospective study was conducted to detect the positive expression rate of CELF1 protein in PTC and paracancerous tissues by immunohistochemistry;CELF1 low expression cell lines were established by small molecular RNA,and the blank control group,the negative control group and the experimental group were set up.The expression of CELF1 mRNA was measured,and the proliferation and invasion of the three groups of cells were detected by CCK method and Transwell chamber test respectively.Result:The positive expression rate of CELF1 protein in PTC tissues was 75.58% (65/86),which was significantly higher than 39.53% (34/86) in paracancerous tissues (χ^(2)=22.87,P<0.01).The expression of CELF1 mRNA in PTC cells of the experimental group (1.06±0.09) was significantly lower than those of the blank control group (2.21±0.16) and the negative control group (2.33±0.13) (F=154.76,P<0.01).Compared with the blank control group and the negative control group,the proliferation ability of the experimental group was significantly decreased after 1 and 2 d of cell culture (F=47.95,56.72,P<0.01).Compared with the blank control group (122.86±15.34) cells and the negative control group (120.57±13.54) cells,the number of invasive cells in the experimental group (84.13±10.04) cells was also significantly less (F=60.24,P<0.01).Conclusion:The expression of CELF1 is high in PTC tissue,and the specific reduction of CELF1 mRNA expression in PTC cells can inhibit cell proliferation and reduce cell invasion.
