美洲大蠊PCR鉴别方法研究

A Method of Identification of Periplaneta Americana

目的探索建立美洲大蠊DNA鉴别方法。方法设计美洲大蠊特异性引物,对模板DNA进行PCR扩增,琼脂糖凝胶电泳法对美洲大蠊及其不同属种蜚蠊进行鉴别。结果采用PCR法首次对美洲大蠊及其不同属种蜚蠊进行鉴别,并对退火温度、循环次数考察,确定其最优条件,对不同Taq酶、不同PCR仪进行耐用性验证,结果均符合要求。结论首次发现并建立美洲大蠊DNA鉴别方法,专属性强,对原料药的掺杂混伪品情况进行严格控制。

Objective To establish a DNA identification method for Periplaneta Americana.Methods The specific primers of Periplaneta Americana were designed。The template DNA was amplified by PCR.Periplaneta Americana and blattariae of different species were identified via agarose gel electrophoresis.Results A PCR method was used to identify Periplaneta Americana and blattariae of different species.The annealing temperature and the number of times of cycling were investigated to determine the optimal conditions.The durability verification of different Taq enzymes and different PCR instrument suggested that the results met the requirements.Conclusion This is the first time that a DNA identification method has been established for Periplaneta Americana.This method has a strong specificity and can exercise rigorous control over the doping and mixing of raw materials.
