体外3T3细胞中性红摄取试验评价复方茜草增色液的光毒性作用

In vitro Neutral Red Uptake Assay of 3T3 Cells for Assessment of Phototoxicity of Compound Qiancao Zengse Liquid

目的复方茜草增色液是根据我院临床经验方制备的搽剂,对治疗白癜风有很好的疗效,本研究采用体外BALB/c 3T3细胞中性红摄取试验,评价复方茜草增色液是否具有光毒性。方法通过对比BALB/c 3T3细胞中性红摄取试验中阳性对照(盐酸氯丙嗪)与受试样品在有光照及无光照两种条件下的细胞活性、IC_(50)值、光刺激因子(PIF)和平均光效应(MPE),对样品产生光毒性的情况进行预测。结果阳性对照盐酸氯丙嗪出现明显的光毒性反应,而复方茜草增色液在有光照及无光照条件下,其PIF值均为*1,MPE值为-0.088<0.1,光毒性反应为阴性。结论表明复方茜草增色液在本试验条件下对BALB/c 3T3细胞无光毒性产生,安全性良好。

Objective To evaluate the possible skin phototoxicity of compound qiancao zengse liquid,which is a liniment prepared based on the clinical experience of our hospital.Methods The phototoxicity of BALB/c 3T3 cells was predicted by comparing the cell activity,IC_(50) value,PIF and MPE of compound Qiancao Zengse liquid with the positive control(CPZ)via the neutral red uptake test with and without light.Results The positive control CPZ manifested significant phototoxicity,but the PIF value of compound qiancao zengse liquid with or without UV exposure was,1,the MPE value was-0.088<0.1,and the phototoxicity was negative.Conclusion Compound Qiancao Zengse liquid is safe and causes no phototoxicity to BALB/c 3T3 cells under the conditions adopted in this experiment.
