茜素红S对鲈鲤幼鱼肌肉抗氧化指标的影响 被引量:2

摘要 在水温(14±0.5)℃下,将体长(5.89±0.62)cm、体质量(1.48±0.39)g的鲈鲤Percocypris pingi幼鱼放入19 cm×13 cm×9 cm的塑料箱中,用浓度为0 mg/L、40 mg/L、80 mg/L、120 mg/L、160 mg/L、200 mg/L和240 mg/L的茜素红S(ARS)溶液浸泡24 h后取鱼样一次(0 d),将剩余鲈鲤幼鱼转入盛有清水的塑料箱中投喂,暂养5 d和10 d时测定鱼体肌肉中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)含量,研究ARS标记对鲈鲤幼鱼肌肉抗氧化指标的影响。结果显示:浸泡24 h(0 d)后SOD和CAT活性随ARS浓度升高呈抛物线型剂量效应体系,表现为低促高抑;在40 mg/L浓度时开始诱导GSH-Px活性,整体呈不规则变化;MDA含量随ARS的浓度先降低后升高,200 mg/L浓度组SOD、CAT和GSH-Px酶活力和MDA含量最高;浸泡5~10 d后,ARS的浓度为40~120 mg/L时,肌肉抗氧化酶活性和MDA含量都能恢复到正常水平,随着ARS的浓度提高160~240 mg/L浓度组的抗氧化酶活性不同程度地低于对照组,MDA含量分别在第5 d和第10 d显著低于和高于对照水平,说明该浓度范围内,ARS浸泡对鲈鲤肌肉造成了不可逆的氧化损伤。本实验结果表明:小于160 mg/L浓度的ARS浸泡鲈鲤未造成氧化损伤。体长4.27~6.51 cm的鲈鲤幼鱼推荐使用100~150 mg/L浓度的ARS进行浸泡标记。 Juvenile Percocypris pingi with body length of(5.89±0.62)cm and body weight of(1.48±0.39)g were reared in a 19cm×13 cm×9 cm plastic box,and exposed to alizarin red S(ARS)solution at a concentration of 0 mg/L,40 mg/L,80 mg/L,120mg/L,160 mg/L,200 mg/L,and 240 mg/L for 24 h and then fish samples were taken once(0 d),and the remaining juveniles were transferred to a plastic box filled with clear water at water temperature of(14±0.5)℃.Then the activities of superoxide dismutase(SOD),catalase (CAT)and glutathione peroxidase(GSH-Px)and the content of malondialdehyde(MDA)were determined in fish muscles at 5 d and 10 d to the effect of ARS labeling on the muscle antioxidant indices of P.pingi juveniles.The results showed that the juveniles soaked in 24 h had the SOD and CAT activities with parabolic dose-effect relationship with the increase in ARS concentration,with promotion of the antioxidant enzyme activity at low concentration of ARS,and the opposite at the high concentration.GSH-Px activity was shown to be induced in the 40 mg/L concentration group,with irregular changes overall,and MDA content was first found to be decreased and then increased with the elevated ARS concentration,and with the maximal activities of SOD,CAT,and GSH-Pxand MDA content in the 200 mg/L concentration group.The antioxidant enzyme activity and MDA content was recovered to normal levels at 40~120 mg/L in the animals soaked for 5~10 d,with the increase in the concentration,the lower antioxidant enzyme activity in the 160~240 mg/L concentration group than that in the control group,and the significantly lower MDA content than the control group on the 5th day(P<0.05),and significantly higher than that in the control group on the 10th day(P<0.05),indicating that ARS soaking led to irreversible oxidative damage to the fish muscles.The findings indicated that ARS concentration of less than160 mg/L did not cause oxidative damage in P.pingi soaked in ARS.It is recommended to use 100~150 mg/L ARS for the juveniles with body length of 4.27~6.51 cm to immerse them as a mark.
