头颈部鳞状细胞癌铁死亡相关lncRNA预后模型的建立与验证 被引量:1

Establishment and validation of ferroptosis related lncRNA prognosis model in head and neck squamous cell carcinoma

摘要 目的研究头颈部鳞状细胞癌(head and neck squamous cell carcinoma,HNSCC)铁死亡相关的长链非编码RNA(long non-coding RNA,lncRNA)并构建相关的HNSCC预后模型。方法癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库获取HNSCC的RNA高通量转录组数据,FerrDb数据库获取和铁死亡相关基因,并基于HNSCC中铁死亡相关差异表达的lncRNA构建HNSCC预后的多lncRNA模型。结果共获得246个HNSCC铁死亡相关基因和1328个铁死亡相关lncRNA;通过单因素Cox回归分析、LASSO回归及多因数Cox回归分析共筛选得到72个具有独立预后意义的lncRNA构建HNSCC预后模型(均P<0.05)。Kaplan-Meier生存曲线显示,高风险组和低风险组的总生存率比较,差异具有统计学意义(P<0.01)。1、2和3年受试者操作特性(receiver operating characteristic,ROC)曲线下面积分别为0.741、0.739和0.738。结论通过生物信息学方法筛选出72个影响HNSCC预后的铁死亡相关lncRNA,并成功构建预后风险模型,这72个铁死亡相关的lncRNA可能是HNSCC预后的分子生物标志物和治疗靶点。 Objective To investigate the long non-coding RNAs(lncRNAs)related to ferroptosis in head and neck squamous cell carci-noma(HNSCC)and to construct a prognosis model for HNSCC.Methods The Cancer Genome Atlas(TCGA)database was used to obtain transcriptional data of HNSCC,and FerrDb database was used to acquire genes related to ferroptosis.lncRNA model for the prognosis of HNSCC was constructed based on differentially expressed lncRNAs in HNSCC.Results Totally 246 ferroptosis related genes and 1328 ferroptosis related lncRNAs in HNSCC were obtained.A total of 72 lncRNAs with independent prognostic significance were screened by univariate Cox regression analysis,Lasso regression and multivariate Cox regression analysis to construct a prognostic model for HNSCC(all P<0.05).Kaplan-Meier survival curve showed that there was statistically significant difference in overall survival between the high-risk group and the low-risk group(P<0.01).The areas under the receiver operating characteristic(ROC)curves of 1,2 and 3 years were 0.741,0.739,and 0.738,respectively.Conclusions Through bioinformatic methods,72 ferroptosis related lncRNAs affecting the prognosis of HNSCC were screened,and a prognostic model was successfully constructed.These 72 ferroptosis associated lncRNAs may be molecular biomarkers for HNSCC prognosis and therapeutic targets for HNSCC.
