丹参-檀香不同配比抗心肌缺血的作用

The Effect of Different Ratio of Salvia Miltiorrhiza and Sandalwood on Myocardial Ischemia-reperfusion Injury

摘要 目的考察丹参-檀香不同配比对心肌缺血再灌注损伤大鼠的治疗作用。方法将60只SD大鼠随机分为6组:假手术对照组、模型组、阳性对照组、丹参-檀香给药5 g∶5 g组(a组)、丹参-檀香给药10 g∶5 g组(b组)、丹参-檀香给药15 g∶5 g组(c组),每组10只,雌雄各半,灌胃给药。给药14 d,在末次给药1 h后,制备大鼠心脏冠状动脉左前降支结扎-松解的心肌缺血再灌注损伤模型,取血及心肌组织。通过测定心肌梗死率,血清中的肌酸激酶(CK)、乳酸脱氢酶(LDH)含量以及心肌组织中的超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)的含量,优选丹参-檀香治疗心肌缺血的最佳配比。结果与假手术组相比,模型组大鼠的心肌梗死率、CK、LDH及MDA水平显著升高(P<0.01),GSH-Px和SOD水平显著降低(P<0.01),说明建模成功;与模型组相比,给药a、b、c组的心肌梗死面积、LDH、CK及MDA水平明显降低(P<0.01),除给药a组的GSH-Px水平升高(P<0.05)外,其余给药组GSH-Px水平显著升高(P<0.01)。其中,b组和c组大鼠的检测数据结果差异无统计学意义(P>0.05),且2组均优于a组。结论本着治疗效果结合经济效益的原则,最终选择b组配比,即丹参:檀香为10g∶5g为最佳配比。 Objective To investigate the optimal ratio of Salvia miltiorrhiza and sandalwood in the treatment of myocardial ischemia-reperfusion injury in rats.Methods Sixty SD rats were randomly divided into 6 groups:sham operation control group,model group,positive control group,Salvia miltiorrhiza-Sandalwood administration 5 g∶5 g group(group a),Salvia miltiorrhiza-Sandalwood administration 10 g∶5 g group(group b),Salvia miltiorrhiza-Sandalwood administration 15 g∶5 g group(group c),10 rats in each group,half male and half female,intragastric administration.After 14 days of administration,1 h after the last administration,the myocardial ischemia-reperfusion injury model of left anterior descending coronary artery ligation-release was prepared,and blood and myocardial tissue were collected.By measuring the area of myocardial infarction,the content of creatine kinase(CK)and lactate dehydrogenase(LDH)in serum,and the content of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in myocardial tissue,the optimal ratio of Salvia Miltiorrhiza and Sandalwood in the treatment of myocardial ischemia was optimized.Results Compared with the sham operation group,the myocardial infarction area,CK,LDH and MDA levels in the model group were significantly increased(P<0.01),and the levels of GSH-Px and SOD were significantly decreased(P<0.01),indicating that the modeling was successful.Compared with the model group,the myocardial infarction area,LDH,CK and MDA levels in the administration groups a,b and c were significantly decreased(P<0.01).In addition to the increase of GSH-Px level in the administration group a(P<0.05),the GSH-Px levels in the other administration groups were significantly increased(P<0.01).Among them,there was no significant difference between group b and group c,and both groups were better than group a.Conclusion In line with the principle of economic and effective treatment and economic benefits,the best choice is group B whose Salvia miltiorrhiza sandalwood ratio is 10 g∶5 g.Results of this experiment can provide reference for the ratio and method of Salvia miltiorrhiza sandalwood pair in clinical use.
