唐德链霉菌产尼可霉素Z的菌种生长特性及发酵液预处理条件优化

摘要 目的 以唐德链霉菌为生产菌株,进行微生物发酵生产尼可霉素Z,研究该菌种生长特性和发酵料液预处理条件。方法 通过微生物发酵培养,摸索菌种生长和发酵特性。对发酵料液进行不同pH调节,采用不同溶剂进行萃取,得到发酵液预处理条件,并通过高效液相色谱法进行含量检测。结果 唐德链霉菌单菌落平板以采用无微量元素的ISP2培养基,平板pH控制在7.0~7.5,种子培养控制在30 h为宜,前体尿嘧啶和乙酰胺能促进发酵产素。可用草酸或盐酸将发酵料液pH控制在2.0左右,用20%~50%甲醇溶液可完全萃取发酵液中的尼可霉素。结论 通过对唐德链霉菌的菌种生长特性研究,以及对其发酵产尼可霉素发酵液预处理条件的摸索为尼可霉素Z产业化大生产奠定了基础。。 Objective Nikkomycin Z is the fermentation production of Steptomyces tendae.This study aimed to research the growth characteristics of Steptomyces tendae and the pretreatment conditions of the fermentation liquid.Methods The growth and fermentation characteristics of Steptomyces tendae was studied by microbial fermentation culture.The pretreatment conditions of fermentation liquid was obtained through the pH was adjusted by oxalic acid or hydrochloric acid and the fermentation broth was extracted with different solvent.The titer of nikkomycin Z was detected by HPLC.Results It was advisable to use ISP2 medium without trace element.The pH of ISP2 medium should be adjusted between 7.0 and 7.5.The seed should be controlled within 30 h.The precursor uracil and acetamide could promote the production of Nikkomycin Z.Fermentation liquid can be used to adjust pH at about 2.0 by oxalic acid or hydrochloric acid.Nikkomycin Z of fermentation can be completely extracted by 20%~50% methanol solution.Conclusion It laid a foundation to achieve the industrialized production of nikkomycin by the study on the characteristics of Steptomyces tendae and the pretreatment conditions of the Nikkomycin Z fermentation liquid.
