高乳糖饮食叠加水平台法脾虚证模型研究与评价 被引量:4

摘要 目的:在高乳糖饲喂加水平台法所致脾虚证模型基础上,叠加饥饱失常因素,完善模型,并观察检测模型各时间点相关指标变化情况,为后续研究提供数据。方法:采用随机数字表法将42只SD大鼠分为7组,其中正常组12只,模型SD大鼠分为造模2周组、造模3周组、造模4周组,每组6只,造模2周停造1周组、造模3周停造1周组、造模4周停造1周组,每组4只。监测大鼠一般体征、体质量、粪便含水量、抓力。取材后检测血清D-木糖、胃蛋白酶原、胃促生长素、三酰甘油(TG),肌肉腺苷三磷酸、糖原,血小板、红细胞与小肠苏木精-伊红染色。结果:与正常组比较,各造模组体质量降低(P<0.05),造模3周组差值最大;抓力降低(P<0.05),造模2周组差值最大;粪便含水量增加(P<0.05),造模2周组差值最大;造模2周组血清D-木糖水平明显下降(P<0.001);造模3周停造1周组血清胃促生长素水平明显降低(P<0.05);造模2周停造1周组和造模3周组血清TG水平增加(P<0.05);造模2周停造1周组肌肉糖原水平明显减少(P<0.05);造模3周组血小板计数明显减少(P<0.001);各造模组红细胞计数明显增加(P<0.01);各造模组均出现小肠生理结构破坏,造模3周组最显著。结论:增加饥饱失常因素的40%高乳糖饮食加水平台法可成功建立大鼠脾虚证模型,2周时“脾主运化”功能降低明显,3周时脾虚证体征和“脾主统血”功能降低明显,且停止造模后模型不能维持。 Objective:On the basis of the spleen deficiency model caused by high lactose diet and water platform method,the factors of hunger and satiety were combined to improve the spleen deficiency model.Related indicators of the model at each time node were observed and tested to provide data for follow-up research.Methods:A total of 42 SD rats were divided into 7 groups by random number table,including normal group(n=12).The modeled SD rats were divided into 2-week modeling group,3-week modeling group,and 4-week modeling group,with 6 in each group,and 2-week modeling+1-week stop group,3-week modeling+1-week stop group,4-week modeling+1-week stop group,with 4 in each group.The general signs,body mass,fecal water content,and grasping force of the rats were monitored.Samples were collected to detect serum D-xylose,pepsinogen,ghrelin,triglyceride(TG),muscle adenosine triphosphate(ATP),glycogen,platelets,red blood cells and hematoxylin-eosin stained small intestine.Results:Compared with the conditions in normal group,the body mass and grasping force were reduced in modeling groups(both P<0.05),and the 3-week modeling group and the 2-week modeling group had the largest difference in the two indicators,respectively;the fecal water content was increased(P<0.05),and the 2-week modeling group had the largest difference.The serum D-xylose level in the 2-week modeling group and the concentration of serum ghrelin in the 3-week modeling+1-week stop group were lower than those in the normal group(P<0.001,P<0.05).The 2-week modeling+1-week stop group and 3-week modeling group had elevated serum TG level(P<0.05),and the 2-week modeling group+1-week stop and the 3-week modeling group had lowered content of muscle glycogen and platelet count,respectively(P<0.05,P<0.001)as compared with the conditions in the normal group.Additionally,the count of red blood cells in each modeling group was increased(P<0.01),and the physiological structure of small intestine was destroyed,with the 3-week modeling group the most significant.Conclusion:High lactose diet(40%)and water platform method combined with hunger and satiety could successfully induce a rat model of spleen deficiency syndrome.Specifically,the function of“spleen governing transport and transformation”was reduced significantly at 2 weeks;the signs of spleen deficiency and the function of“spleen governing blood”were decreased significantly at 3 weeks.The model failed to be maintained after the modeling was stopped.
