摘要
目的研究银屑胶囊治疗银屑病可能的通路及机制。方法利用中药系统药理学数据库与分析平台(TCMSP)检索银屑胶囊的有效成分和靶标。并通过GeneCards数据库、OMIM数据库、PharGKB数据库、DrugBank数据库,以银屑病为关键词筛选总结出关联度高的疾病靶点;利用VennPainter软件将银屑胶囊靶点与疾病靶点进行交叉,筛选出两者的共同靶点。将银屑胶囊中的化合物、银屑病及其共同靶标蛋白上传至Cytoscape软件,生成“药物-共同靶标-疾病”网络图,探讨银屑胶囊的药理作用机制。利用String蛋白互作数据库和Cytoscape软件分析常见靶蛋白之间的相互作用,完成Gene Ontology(GO)富集分析和Reactome富集分析。结果以口服生物利用度(OB)≥30%、药物相似度(DL)≥0.2为标准,筛选出银屑胶囊28个有效成分,117个靶点,共鉴定出银屑病高相关度靶点766个。药物和疾病共同靶点88个。GO富集分析包括89个生物学过程结果,306个分子功能结果和78个细胞组成结果。Reactome富集分析共83个结果,共同靶点的功能主要表达在γ-氨基丁酸(GABA)受体、NMDA等通路上。结论银屑胶囊对银屑病的治疗作用机制主要集中在对γ氨基丁酸受体通路和NMDA通路的调控方面。
Objective Network pharmacology was used to explore the mechanism of Yinxie Capsule(银屑胶囊)in the treatment of psoriasis.Methods The active compounds and targets of Yinxie Capsule were searched by TCMSP,the pharmacological database of Chinese medicine system.And through the GenCards database,OMIM database,PharGKB database,DrugBank database with psoriasis as the key words to screen and summarize the disease targets with high correlation degree;VennPainter software was used to intersect the targets of Yinxie Capsule compound and disease targets,and the common targets of the two were screened out.The compounds,psoriasis and their common target proteins in Yinxie Capsule were uploaded to Cytoscape software to generate a network diagram of“drug-common target-disease”to explore the pharmacological mechanism of action of Yinxie Capsule.The Gene Ontology(GO)enrichment analysis and Reactome enrichment analysis were further completed by using the String protein interaction database and Cytoscape software to analyze the interactions between common target proteins.Results Based on the criteria of oral bioavailability(OB)≥30%and drug likeness(DL)≥0.2,a total of 28 active compounds of Yinxie Capsule were screened out,117 targets,and total of 766 targets with high correlation degree in psoriasis were identified.There were 88 common targets for drugs and diseases.GO enrichment analysis included 89 results of biological process,306 results of molecular function and 78 results of cell composition.Reactome enrichment analysis included 83 results,indicating that the functions of common targets were mainly expressed in the pathways such asγ-amino butyric acid(GABA)receptor and NMDA.Conclusion Based on network pharmacology,this study clarified the therapeutic effect of Yinxie Capsule on psoriasis,and the mechanism mainly focused on the regulation of GABA receptor pathway and NMDA pathway,which provided a basis for clinical application and research.
