紫檀芪通过PI3K/Akt通路对脓毒症大鼠急性肺损伤的保护作用

Protective effect of pterostilbene on acute lung injury in septic rats through PI3K/Akt pathway

目的 分析紫檀芪(PTS)对脓毒症大鼠急性肺损伤的保护作用及其可能作用机制。方法 将大鼠30只随机分为假手术组、脓毒症组和PTS组,每组10只,脓毒症组和PTS组(25 mg/kg)均通过盲肠结扎穿刺术建立大鼠脓毒症模型,24 h后处死大鼠;TUNEL染色法检测肺组织细胞凋亡,ELISA法测定血清肿瘤坏死因子(TNF-α)、白介素-6(IL-6)和单核细胞趋化蛋白1(MCP-1)含量,Western blot检测肺组织中p-PI3K和p-Akt的表达。结果 与假手术组比较,脓毒症组肺组织TUNEL阳性细胞数、血清TNF-α、IL-6、MCP-1含量均升高(P<0.05),肺组织p-PI3K和p-Akt蛋白含量均下降(P<0.05);而与脓毒症组比较,PTS组肺组织TUNEL阳性细胞数、血清TNF-α、IL-6、MCP-1含量均下降(P<0.05),肺组织p-PI3K和p-Akt蛋白含量均增加(P<0.05)。结论 PTS通过PI3K/Akt途径抑制脓毒症所致大鼠肺损伤的凋亡和炎性反应,为脓毒症所致肺损伤的药物治疗提供候选。

Objective To analyze the protective effect of pterostilbene(PTS) on acute lung injury in septic rats and its possible mechanism.Methods Thirty rats were randomly divided into sham operation group, sepsis group and PTS group,10 in each group, the sepsis group and the PTS group(25 mg/kg) were all established by cecal ligation and puncture, after24 hours, the rats were sacrificed;The apoptosis of diaphragm tissues was evaluated using TUNEL assay, the content of TNF-α, IL-6 and MCP-1 in lung specimen were measured by ELISA assa, Western blot were used to explore the levels of p-Akt and p-PI3K in lung tissues.Results Compared with the sham operated group, the number of TUNEL-positive cells,TNF-α, IL-6 and MCP-1 contents in the lung tissue of the sepsis group were significantly increased(P< 0. 05), while p-PI3K and p-Akt protein content decreased(P< 0. 05);Compared with the sepsis group, the number of TUNEL-positive cells, the contents of TNF-α, IL-6 and MCP-1 in the lung tissue of the PTS group were significantly decreased(P< 0. 05), while p-PI3K and p-Akt protein content were significantly increased(P< 0. 05).Conclusion PTS inhibits the apoptosis and inflammatory response of sepsis-induced lung injury in rats through the PI3K/Akt pathway, providing a candidate for drug therapy of sepsis-induced lung injury.
