中、意蜂蜂毒对小鼠耳肿胀炎性抑制作用的比较 被引量:1

Inflammation inhibitory effects of bee venom from Apis cerana cerana or Apis mellifera ligustica on ear swelling in mice

摘要 [目的]蜂毒由工蜂的毒腺合成,螫刺时由螫针排出,具有良好的生物学活性和药理活性。本研究旨在探讨和比较中蜂蜂毒和意蜂蜂毒对小鼠急性耳肿胀的影响及其潜在作用机理。[方法]选用体重30 g左右的健康昆明小鼠,按照试验要求随机分组。采用二甲苯诱导小鼠急性耳肿胀模型,比较蜂毒作用前后小鼠耳肿胀度和肿胀率,ELISA法检测血清中炎症介质PGE2、TNF-α和核因子NF-κB的含量,qRT-PCR法检测炎性因子IL-1β、IL-4和IL-6的表达水平。[结果]与阴性对照组相比,模型组小鼠耳肿胀程度和肿胀率显著增加,血清中TNF-α、PGE2和NF-κB含量均显著升高,耳组织中IL-1β、IL-6的基因表达水平显著升高,IL-4的基因表达水平显著降低。与模型组相比,使用不同剂量蜂毒处理后均能显著减轻小鼠耳肿胀度和肿胀率(P<0.05),并使得小鼠血清中升高的TNF-α、PGE2和NF-κB含量显著降低(P<0.05),使小鼠耳组织中IL-1β和IL-6的表达量显著降低,而IL-4的表达量显著升高(P<0.05)。试验结果还发现,中蜂蜂毒和意蜂蜂毒对小鼠耳肿胀及炎症介质、核因子和炎性因子表达量的影响无显著差异(P>0.05)。[结论]蜂毒可显著减轻二甲苯所致小鼠的急性炎症,对急性炎症的保护机制可能与调节炎症介质和炎症因子释放有关。中蜂蜂毒的抗炎效果略低于意蜂蜂毒,但二者间差异不显著。 [Objective]The honey bee venom is a well-known biologically and pharmacologically active compound synthesized by the venom glands of workers and discharged from the sting apparatus when stinging.The purpose of this study was to investigate and compare the effects and the potential mechanism of bee venom(BV)from Apis cerana cerana and Apis mellifera ligustica on acute ear swelling in mice.[Method]Healthy Kunming mice weighing about 30 g were randomly selected and divided into different groups according to the experimental requirements.The acute ear swelling model induced by xylene was used to compare the degree of ear swelling and swelling rate of mice before and after BV application.The levels of PGE2,TNF-αand NF-κB in serum were detected by ELISA assay and the expression levels of IL-1β,IL-4 and IL-6 mRNA were detected by qRT-PCR.[Result]The results showed that compared with the negative control group,the ear swelling degree and swelling rate of the mice in the model group was significantly increased.The levels of serum TNF-α,PGE2 and NF-κB,were significantly increased too.The gene expression levels of IL-1βand IL-6 were significantly increased,but that of the IL-4 was significantly decreased(P<0.05).Compared with the model group,the BV treatment groups with different concentration significantly reduced the degree and rate of ear swelling induced by xylene in mice(P<0.05).The levels of TNF-α,PGE2 and NF-κB in the serum were significantly decreased,and the gene expression levels of IL-1βand IL-6 in the tissue of ear were significantly decreased but the IL-4 was significantly increased(P<0.05).In addition,the results also indicated that there was no significant difference of BV from A.cerana cerana or A.mellifera ligustica on the effects of inhibiting ear swelling and the expression of inflammatory mediators,nuclear factors and inflammatory factors(P>0.05).[Conclusion]The results indicate that BV can significantly reduce the acute inflammation induced by xylene,and its protective mechanism to acute inflammation may be related to regulating the release of inflammatory mediators and inflammatory factors.Although the anti-inflammatory effect of BV of A.cerana cerana was lower than that of A.mellifera ligustica,the difference was not significant.
