室旁核HO-1对高盐性高血压PI3K/Akt通路的影响 被引量:1

摘要 目的:通过影响高盐诱导的高血压大鼠室旁核(paraventricular nucleus, PVN)HO-1表达,观察PVN中PI3K-p85和Akt的蛋白表达变化,探讨室旁核HO-1对高盐性高血压PI3K/Akt通路的影响。方法:选取成年雄性Dahl盐敏感大鼠40只,随机平均分成4组,即NS+PVN vehicle组、NS+PVN ZnPP Ⅸ组、HS+PVN vehicle组和HS+PVN ZnPP Ⅸ组。分别饲喂正常饮食(NS)或高盐饮食(HS),双侧PVN插管持续微量注射ZnPP Ⅸ,采用尾动脉无创测量平均动脉压(MAP),采用ELISA法检测血浆NE,采用Western blotting法检测PVN中PI3K-p85、p-Akt、t-Akt和HO-1的蛋白表达。结果:与饲喂NS大鼠相比,饲喂HS大鼠MAP和血浆NE水平明显升高,室旁核PI3K-p85和p-Akt蛋白表达明显升高(P<0.05);与HS+PVN vehicle组相比,HS+PVN ZnPP Ⅸ组MAP和血浆NE水平明显升高,室旁核PI3K-p85和p-Akt蛋白表达明显升高(P<0.05)。结论:室旁核PI3K/Akt信号通路激活可促进高盐性高血压的外周交感神经活动和血压增高,室旁核HO-1可能通过抑制PI3K/Akt信号通路而起到降低外周交感神经活动和血压的作用。 Objective: To investigate the effect of HO-1 in paraventricular nucleus on PI3 K/Akt pathway in high-salt induced hypertension. Methods: Dahl salt sensitive rats were randomly divided into four groups(n=10): NS + PVN vehicle group, NS + PVN ZnPP Ⅸ group, HS + PVN vehicle group and HS + PVN ZnPP Ⅸ group. High salt diet(HS) and normal salt diet(NS) were fed respectively. The blood pressures of rats were measured through the caudal arteries. The inhibitors of HO-1(ZnPP Ⅸ) were microinjected in the bilateral PVN of rats by micro osmotic pumps. The plasma noradrenaline(NE) levels were detected by ELISA. And the protein expressions of PI3 K-p85, p-Akt, t-Akt and HO-1 in the PVN were detected by western blotting. Results: Compared with the NS groups, the levels of the blood pressure and the plasma NE and the expressions of PI3 K-p85 and p-Akt in the PVN were significantly increased in the HS groups(P<0.05). Compared with the HS + PVN vehicle group, the levels of the blood pressure and the plasma NE and the expressions of PI3 K-p85 and p-Akt in the PVN were significantly increased in the HS + PVN ZnPP Ⅸ groups(P<0.05). Conclusion: Activation of PI3 K/Akt signaling pathway in the PVN may promote peripheral sympathetic nervous system activity and increase blood pressure in the high-salt induced hypertension. The expression of HO-1 in the PVN affects the blood pressure in high salt hypertension, which may be related to PI3 K/Akt pathway.
