摘要
目的 从浙江温岭红树林秋茄中筛选分离获得乙酰胆碱酯酶抑制活性菌株,并对较强活性菌株WHSL-08进行菌种鉴定。方法 以浙江省台州温岭市的红树林秋茄样品为材料,进行海洋真菌分离,采用Ellman法对分离到的菌株进行乙酰胆碱酯酶抑制活性筛选。通过菌株培养性状、形态特征及ITS r DNA基因测序等方法,对菌株WHSL-08进行菌种分类鉴定。结果 本研究从秋茄根际土壤、根、茎和叶中共分离获得41株海洋真菌,经测定有16株具有乙酰胆碱酯酶抑制活性,活性菌株率达39.02%,其中根际土壤分离的菌株数最多,活性菌株率达50%。活性菌株中WHSL-08活性最强,经进一步菌种鉴定确定该菌株为Isaria javanica。结论 浙江温岭秋茄根系土壤、根、茎和叶中均可筛选到乙酰胆碱酯酶抑制活性菌株,根系土壤中的活性菌株率较高。活性最强菌株WHSL-08经鉴定为Isaria javanica,其代谢产物值得进一步研究开发。
OBJECTIVE To identify the higher antiacetylcholinesterase activity of the WHSL-08,fungi strains were screened and isolated from mangrove( kandelia candel).METHODS In this paper,marine fungus strains were isolated from the kandelia candel in Wenling,Taizhou,Zhejiang Province.The acetylcholinesterase inhibitory activity of each strain was detected using Ellman method.The strain WHSL-08 with stronger activity was identified by strain culture characteristics,morphological characteristics and ITS r DNA gene sequencing.RESULTS A total of 41marine fungus strains were isolated from rhizosphere soil,roots,stems and leaves of kandelia candel.Furthermore,16of 41 strains with inhibitory activity were obtained,and the total positive rate was 39.02%.The most fungi strains and fungi strains with antiacetylcholinesterase activity were isolated from rhizosphere soil.The strain WHSL-08 with the highest antiacetylcholinesterase activity was identified as Isaria javanica.CONCLUSION The antiacetylcholinesterase active strains could be screened and isolated from the rhizosphere soil,roots,stems and leaves of kandelia candel in Wenling,Zhejiang Province.The active strains from rhizosphere soil were more than other tissues.Among the active strains,WHSL-08 Isaria javanica has the highest inhibitory activity,and its active metabolites are worth to be studied further.
